Objective:To investigate the association between the polymorphisms of MRAS gene rs146298087,rs138024847,rs3821696,rs1002766,rs1199333,rs1199332,rs40593 and rs229325 and the susceptibility to obesity in children aged 6-13 years in Shawan County,Xinjiang,it also provides a potential therapeutic target for the prevention and treatment of obesity.Methods:A survey was conducted on the current situation of a primary school in Shawan City,Xinjiang,with 100 children randomly selected from the obesity group and 100 children randomly selected from the control group.4 ml of fasting venous blood was sampled,and the polymorphisms of MRAS genes rs146298087,rs138024847,rs3821696,rs1002766,rs1199333,rs1199332,rs40593 and rs2293252 were detected by high-throughput sequencing,and the relationship between MRAS genes and childhood obesity susceptibility was analyzed.Results:1)In the constructed dominant model,the polymorphism of the MRAS gene rs40593(A/A,A/G+G/G)was correlated with the occurrence of obesity in children aged 6-13.The likelihood of obesity in individuals carrying the A/G+G/G genotype was significantly increased(OR=1.821,95%CI:1.021-3.248).Further gender analysis revealed a correlation between the rs40593(G/A)polymorphism and the occurrence of obesity in boys.When using the G allele as a reference,individuals carrying the A allele were significantly less likely to develop obesity(OR=0.343,95%CI:0 144-0 819);The polymorphism of rs40593(GG/AG/AA)is associated with the occurrence of obesity in boys.2)The rs3821696(G/C)polymorphism of the MRAS gene is associated with the occurrence of obesity in boys aged 6-13.When the G allele is used as a reference,individuals carrying the C allele are significantly more likely to develop obesity(OR=5.418,95%CI:1.54-19.065),but a correlation between the rs3821696 polymorphism and obesity was not found in girls.3)The polymorphism of the MRAS gene rs1199333(GG/GT/TT)is associated with the occurrence of obesity in boys aged 6-13(P=0.004).In the dominant model,using the TT genotype as the control standard,the likelihood of obesity in individuals carrying the GG/GT genotype was significantly reduced(OR=0.145,95%CI:0.037-0.565),but a correlation between the rs 1199333 polymorphism and obesity was not found in girls.4)The rs 1199332(G/C)polymorphism of the MRAS gene may be associated with the occurrence of obesity in boys aged 6-13 years old(χ2=5.437,P=0.020),when using the G allele as a reference,the likelihood of obesity in individuals carrying the C allele was significantly reduced(OR=0.364,95%CI:0.152-0.87);The polymorphism of MRAS gene rs1199332(GG/CG/CC)is associated with the occurrence of obesity in girls aged 6-13(P=0.035).CONCLUSION:1)In the constructed explicit model,the polymorphism of the MRAS gene rs40593(A/A,A/G+G/G)is associated with the occurrence of obesity in children aged 6-13,and the likelihood of obesity in individuals carrying the A/G+G/G genotype is significantly increased.2)The polymorphisms of MRAS gene rs3821696(G/C),rs1199333(GG/GT/TT),and rs1199332(G/C)are associated with the occurrence of obesity in boys aged 6-13.
