Objecyive: To investigate the curative effect of Xiaoke Jianpi capsule prepared by modern technology and traditional technology in the treatment of type 2 diabetes mellitus(T2DM),to study the effect of Xiaoke Jianpi Capsule on insulin resistance and inflammatory factors of spleen-deficient dampness-T2 DM rats,and to observe the pathological changes of the rats.In order to improve the efficacy and safety of Xiaoke Jianpi capsule,and prepare data for the record of traditional Chinese medicine preparations.Methods: The high-fat diet combined with peritoneal injection of streptozotocin(STZ),overwork + diet deficiency + damp-heat environment were used to establish a T2 DM rat model with spleen deficiency and dampness.The rats were randomly divided into medium and high dose group by modern technology and medium and high dose group by traditional technology.Trough continuous intragastric administration once a day,after 4 weeks of intervention,the general state,diet,water intake,urine volume,body weight and fasting blood glucose of rats were recorded,and the optimal preparation process was optimized by control experiment.Xiaoke Jianpi Capsules were prepared on the basis of traditional technology.The rats were randomly divided into rosiglitazone group,Xiaoke Jianpi capsules low-dose,medium-dose and high-dose groups.The fasting blood glucose(FBG)of rats in each group was detected after continuous drug intervention for 4 weeks.Oral glucose tolerance test(OGTT)was performed at week 5.Glycosylated serum protein(GSP)and serum insulin(INS)were detected and insulin resistance index(HOMA-IR)was calculated.The expression levels of triglyceride(TG),total cholesterol(TC),high density lipoprotein cholesterol(HDL-C),low density lipoprotein cholesterol(LDL-C),C-reactive protein(CRP),interleukin-6(IL-6)and tumor necrosis factor α(TNF-α)were observed by HE staining.Results:There were no significant changes in FBG and body weight between the modern process group and the traditional process group(P > 0.05),but the traditional process improved the three symptoms,hair color,stool and mental state better.Serum index test results showed that compared with normal group,blood glucose and AUC in model group were increased at each time point of FBG and OGTT(P < 0.05).FINS and HOMA-IR were increased(P < 0.05);IL-6,TNF-α and CRP were increased(P < 0.05).Compared with model group,the polydipsia,polyuria and mental state of rats in Xiaoke Jianpi capsule medium and high dose groups were significantly improved(P < 0.05),and the body weight was increased.Blood glucose and AUC were decreased at each time point of FBG and OGTT(P < 0.05).FINS and HOMA-IR decreased(P < 0.05);IL-6,TNF-α and CRP were decreased(P < 0.05).Compared with the model group,the pathological morphology of Xiaoke Jianpi capsules in the low,middle and high three groups was significantly improved.Conclusions: Compared with the modern process,the efficacy of the capsules prepared by the traditional process was more significant in T2 DM with spleen deficiency and dampness.The capsules could effectively improve insulin resistance and inflammation status,and improve body weight and pathological histomorphology of various organs in rats. |
