| Objective:Using network pharmacology approach to predict the targets of action of galangin in the treatment of vitiligo and to explore its mechanism of action;To study the effect of galangin cream on the NLRP3 inflammasome pathway in an animal model of experimental vitiligo.Methods:1.The TCMSP database was used to query the chemical composition of galangin,the active ingredients of galangin were screened and their targets were collected,targets of vitiligo disease were collected through 4 databases,the interaction network between proteins was constructed,the next phase was GO feature enrichment analysis and KEGG pathway enrichment analysis,and molecular docking was used to predict related genes.2.A mouse model of C57 BL/6 vitiligo was established using 10%H2O2induction,and after successful modeling,different concentrations of galangin cream were administered to the mice.The NLRP3 inflammasome pathway-related proteins were detected by western blot and RT-q PCR;the serum was obtained by centrifugation after the removal of eyeballs,and the levels of relevant indicators of the pathway in mouse serum were measured by enzyme-linked immunosorbent assay(ELISA).Results:1.The results of the network pharmacology study showed that the core targets of galangin for vitiligo are tyrosinase(TYR),NOD-like receptor protein 3(NLRP3),inducible nitric oxide synthase(NOS2),tumor necrosis factor(TNF),interleukin-1β(IL-1β),interleukin-18(IL-18),etc.The GO and KEGG The enrichment results showed that the biological processes mainly involved in the treatment of vitiligo with galangin may play a role in cell proliferation and apoptosis,and participate in biological processes such as gene expression,signal transduction,redox,and protein hydrolysis,and the main pathways of action are NOD-like receptor,tyrosine metabolism,HIF-1 and NF-kappa B signaling pathway.2.Pathological sections showed that the epidermis of mice in the normal group had no hyperplasia,while that of mice in the model group showed significant hyperplasia,and the epidermis of mice in the low,medium and high dose groups of galangin cream improved compared with that of the model group.1,NLRP3 and CXCL16 in mice serum(P<0.05);western blot showed that the expression levels of IL-1β,IL-6,ASC,caspase-1,NLRP3,CXCL10 and CXCL16 in mice skin tissues were significantly reduced by low and high doses of galangin cream(P<0.05);RT-q PCR experiments showed that low,medium and high doses of galangin cream significantly reduced the expression levels of caspase-1,IL-1β,IL-6 and CXCL10 m RNA in mouse skin tissues(P<0.05).Conclusion:A preliminary network pharmacological study revealed that the therapeutic effect of galangin on vitiligo has multi-target and multi-pathway characteristics;galangin cream can inhibit epidermal proliferation in vitiligo mice;it exerts its effect on improving vitiligo by inhibiting NLRP3 inflammasome pathway. |
