Objective: To analyze the expression of endothelial cell specific molecules1(ESM-1)in triple negative breast cancer(TNBC),and to explore the effect of ESM-1 on Integrin in the secretion of integrinsβ2 binding to intercellular cell adhesion molecule-1(ICAM-1).Methods: Collect data from the Human Tumor Genome(TCGA)database,analyze the expression of ESM-1 in different tumors,and the relationship between ESM-1 in TNBC and clinical pathological characteristics and prognosis of patients.Data from human tumor genome(TCGA)database were collected to analyze the expression of ESM-1 in different tumors and the relationship between ESM-1 and clinicopathological features and prognosis of patients in TNBC.The tumor exosomes extracted from MDA-MB-231 cell line were co-cultured with DC cells and added with ESM-1.The localization of ITGB2,ESM-1 and ICAM-1 on DC surface was observed by immunofluorescence assay.The expression of ESM-1,ICAM1 and ITGB2 was detected by q RT-PCR.Results: 1)The results of TCGA database showed that the expression of ESM-1 in 127 cases of TNBC cancer tissues was significantly higher than that in adjacent tissues,with statistical significance(P< 0.05).2)Pan-cancer analysis showed that ESM-1 was highly expressed in head and neck squamous cell carcinoma,thyroid cancer,breast cancer,esophageal cancer,stomach cancer,liver cancer,bile duct cell carcinoma,kidney clear cell carcinoma,endometrial carcinoma,bladder urothelial carcinoma,and colorectal cancer.3)Immunofluorescence experiments showed that ITGB2 and ICAM-1 co-localization was significantly reduced after the addition of ESM-1.4)q RT-PCR showed that ITGB2 expression level was up-regulated after ESM-1 was added(P< 0.05).Conclusion: ESM-1is highly expressed in TNBC,and can bind to ITGB2,and then affect the binding between ITGB2 and ICAM-1.Conclusion:ESM-1 is highly expressed in TNBC,and can bind to ITGB2,and then affect the binding between ITGB2 and ICAM-1.
