| Objective: Drawn whole blood samples were collected from 20 children who has right ventricular hypertrophy with high altitude heart disease.The expression of MyoD gene was detected by gene sequencing in the children.On the ground,the clinical significance of MyoD gene expression in children with high altitude heart disease and right ventricular hypertrophy was explored,and the mechanism of right ventricular hypertrophy was further explored.So as to explore the onset and development process of high altitude heart disease at the gene level.Methods: 1.In this study,20 Tibetan children aged 3-14 years who were diagnosed with high altitude heart disease and met the inclusion criteria were selected as the research objects.2.After the subjects were selected,3ml of whole venous blood was drawn from each subjects with anticoagulant tube and stored in the refrigerator at-4℃.DNA was extracted from whole blood samples,and the extracted DNA was stored in a special refrigerator at-50℃,and then the DNA was sent for detection.In the process of testing,the DNA needs to be placed in a special refrigerator at-20 ° C before transport.The extracted DNA was sent to the laboratory for gene sequencing.3.In the laboratory,the entire DNA sequence of the samples will be captured by a dedicated capture technology.4.The quality of the DNA sequence was tested,and the qualified samples were sequenced to detect whether there was MyoD gene expression in the sequence.Result: 1.The MyoD gene is specifically expressed only in skeletal muscle,even in the early embryonic heart.If the expression of MyoD gene is detected in the sample,it is significant.2.In the test of DNA quality,18 A and 2 C were detected in 20 samples,and the samples with quality C were eliminated.18 samples with quality A passed the quality test and were qualified.3.Qualified samples was performed the DNA sequence,and the DNA test results of 18 samples all showed the presence of MyoD sequence.Conclusion: 1.MyoD gene is expressed in Tibetan high altitude heart disease patients with right ventricular hypertrophy.2.MyoD gene may play an important role in the formation of right ventricular hypertrophy in high altitude heart disease. |
