| Objective To explore the regulatory mechanism of Ruangan powder on the pathogenesis of hepatic fibrosis through Hedgehog signal pathway during the occurrence and development of hepatic fibrosis induced by porcine serum,and provide a new entry point for the prevention and treatment of hepatic fibrosis.Methods 48 SPF grade male rats were selected and included in the study.48 SD rats were randomly divided into normal group,model group,Ruangan powder group and Biejia decoction group,with 12 rats in each group.After one week of adaptive feeding of experimental animals,the other three groups,except the normal group,were injected intraperitoneally with inactivated porcine serum for a fixed time of 0.5 m L each time,twice a week,for 8 weeks.After the establishment of the model was confirmed to be successful,the normal group and the model group were given normal saline by gavage,the Ruangan powder group were given water decoction containing crude drug 2.174g/m L by gavage,and the Biejia decoction group were given suspension containing drug powder 0.233g/m L by gavage,the amount of gavage liquid was 6.7 m L/kg,once a day,for 8 consecutive weeks.After operation,the liver tissue of rats in each group was taken;TUNEL method was used to detect the apoptosis of liver tissue in each group;RT-PCR and Western-blot were used to detect the concentration and expression of Smo,Su Fu and Gli2 in liver tissue of rats in each group.Results Hepatic fibrosis rats showed an increase in hepatocyte apoptosis;The administration of Biejia decoction and Ruangan powder can reduce hepatocyte apoptosis in hepatic fibrosis model rats;Compared with the normal group,the relative expressions of Smo,Su Fu,and Gli2 m RNA in the liver tissue of the model group were significantly increased,with statistical significance(P<0.01);Compared with the model group,the relative expression levels of Smo,Su Fu,and Gli2 m RNA in the liver tissues of rats in the Ruangan powder and Biejia decoction groups decreased,and the content of Smo,Su Fu,and Gli2 m RNA in the liver tissues of rats in the Ruangan powder group was the lowest,with statistical significance(P<0.05);Compared with the normal group,the relative expression of Smo,Su Fu,and Gli2 proteins in the liver tissue of the model group increased significantly(P<0.01);Compared with the model group,the relative expression levels of Smo,Su Fu,and Gli2 proteins in the liver tissues of rats in the Biejia decoction group and Ruangan powder group decreased significantly(P<0.01).Conclusion:(1)Ruangan powder can reduce hepatocyte apoptosis in rats with hepatic fibrosis.It has protective effect.(2)Ruangan powder can inhibit the expression of Smo,Su Fu,Gli2 protein and Smo,Su Fu,Gli2 m RNA in the Hedgehog signal pathway,thus inhibiting the hepatic fibrosis in rats. |
