| Purpose:The effect of acupuncture-rehabilitation method on apoptosis and morphological changes of ischemic brain tissue in rats with permanent cerebral ischemia and the regulation of apoptosis-related proteins in ischemic brain tissue were observed and discussed,providing scientific basis for the treatment of acupuncture-rehabilitation method.Method:Male Sprague-Dawley(SD)rats were randomly divided into 5 groups according to the random number table method: sham operation group,model group,acupuncture group,rehabilitation group and acupuncture kang group,with 12 rats in each group(n=12)at 3 time points on day 3,day 7 and day 14,with 180 male SD rats.Among them,the model group,acupuncture group,rehabilitation group and acupuncture-rehabilitation group were treated with modified endovascular embolization,which blocked the blood flow in the middle cerebral artery of rats,thus forming a permanent cerebral ischemia model.No thread plug was needed in the modeling process of sham operation.The sham operation group and model group were not treated after modeling and were fed back in the rat cage.The acupuncture group was treated with head point cluster acupuncture,the rehabilitation group was treated with table running exercise training after modeling,and the acupuncture-rehabilitation group was treated with head point cluster acupuncture combined with table running training.After treatment,neurological function scores of rats were scored by using mNSS scale at 3 preset time points.HE staining was used to observe the morphological changes of ischemic brain tissue.TUNEL staining was used to observe the apoptosis of ischemic brain cells.The expressions of JAK2,STAT3,P-Stat3,BCL-2 and BAX in ischemic brain were analyzed by Western Blot.Results:1.mNSS score: After the successful modeling of permanent cerebral ischemia model and intervention treatment for 3d,7d and 14 d,the scores of all groups were higher than those of the sham operation group,and the difference was statistically significant(p<0.05).3 days after operation,compared with model group,the scores of acupuncture group and rehabilitation group decreased,but the difference was not statistically significant(p>0.05).Compared with model group and treatment group,the score of acupuncture-rehabilitation group was decreased(p<0.05).On day 7and 14 after surgery,compared with the model group,the scores of all treatment groups decreased with the increase of time,and the scores of neurological impairment in the acupuncture-rehabilitation group decreased significantly,with statistical significance(p<0.05).2.HE staining: 3d,7d,14 d after surgery,no significant pathological damage was observed in the brain tissue of rats in the sham operation group,and most nerve cells were orderly in shape and structure,with clear Outlines and centered nuclei.In the rat brain tissue of model group,the number of intact nerve cells was reduced,a large number of nuclei were prolapsed,cell structure was sparse,cell color was deepened,interstitial edema was observed,and cell space was larger.Compared with the model group,the pathological injury degree of brain tissue in the acupuncture-rehabilitation group was reduced,and the related pathological morphology of the acupuncture-rehabilitation group recovered better.3.Apoptosis of ischemic brain tissue: 3d,7d,14 d after surgery,the number of apoptosis in the sham operation group was very small,which was a normal physiological phenomenon.Compared with the sham operation group,the number of apoptosis in the model group was increased(p<0.05).Compared with model group,the number of apoptosis decreased in treatment groups(p<0.05),and the difference was statistically significant.Compared with all treatment groups,the therapeutic effect of acupuncture Kang group was better(p<0.05),and the difference was statistically significant.4.Protein expression of JAK2,STAT3,P-Stat3 and BAX in ischemic brain tissue of ratsCompared with sham operation group,the expressions of JAK2,STAT3,P-Stat3 and BAX protein in brain tissue of model group were significantly increased on day 3,7 and 14 after operation(p<0.05).The difference was statistically significant.Compared with model group,the protein expressions of JAK2,STAT3,P-Stat3 and BAX in acupuncture group,rehabilitation group and acupuncture-rehabilitation group were decreased(p<0.05),and the differences were statistically significant.Compared with acupuncture group and rehabilitation group,the above proteins in acupuncture-rehabilitation group were significantly reduced(p<0.05),and the difference was statistically significant.5.Protein expression of BCL-2 in rat ischemic brain tissueOn day 3,7 and 14 after surgery,the expression of BCL-2 in the model group was significantly decreased compared with the sham group(p<0.05).Compared with model group,the expression of BCL-2 in treatment groups was increased(p<0.05).Among them,the expression level of BCL-2 in brain tissue of rats in acupuncture-rehabilitation group was higher than that in acupuncture group and rehabilitation group,and the difference was statistically significant(p<0.05).Conclusion:1.Acupuncture therapy can reduce the mNSS neurological function score in rats with permanent cerebral ischemia,thus improving the neurological function deficit.2.Acupuncture-rehabilitation method can improve apoptosis of cerebral ischemic tissue in rats to some extent3.Acupuncture-rehabilitation method can reduce the expression of JAK2/STAT3 pathway protein in the brain tissue of rats with permanent cerebral ischemia,and reduce the injury degree of ischemic brain tissue. |
