| Objective There is a complex interaction between the nervous and cardiovascular systems, and previous studies have found that stroke and traumatic brain injury can cause cardiac dysfunction. This experiment explores whether rats with vascular dementia can cause myocardial damage and its possible mechanism,in order to provide a theoretical basis for the early clinical diagnosis and treatment of vascular dementia.Method 24 healthy SPF-grade male SD rats were randomly divided into normal group, model group, sham group according to the random number table method, with 8 rats in each group.2-VO method was used to establish vascular dementia model in the rats of model group. The sham group had the same surgical method as the model group, but bilateral common carotid arteries were not ligated, and the normal group was raised under the same conditions. After the completion of modeling, the rats in each group were carefully raised in a suitable and identical environment for 4 weeks, and then Morris water maze test. The water maze test lasted for one week, and the brain, heart and blood samples of all rats were taken at the sixth week.Subsequently, the pathological changes of the brain and heart were observed by HE staining, and the concentration of norepinephrine(NE),epinephrine(E),arginine vasopressin(AVP)and NO content in serum was detected by ELISA.Results(1)Water maze experimental results: The evasion incubation period of rats in the model group was significantly longer than that of the sham surgery group and the normal group, and the difference was statistically significant(P<0.01).(2)HE staining results: Compared with the sham surgery group and the normal group,it was found that the model group rat cardiomyocytes were irregular in morphology, arranged disorderedly, some cardiomyocytes nuclei were hypertrophy or contraction, inflammatory cell infiltration was visible, and myocardial interstitium was obviously edematous.(3)ELISA test results: The contents of NE,E,AVP and NO in serum were significan. Conclusion Vascular dementia can lead to heart damage. Its mechanism is not caused by a single factor, but may be caused by oxidative stress mediated by sympathetic parasympathetic nerve stimulation. |
