Association Between Exposure To Bisphenol A And Bisphenol B During Pregnancy And Placenta-Derived Progesterone And "Placental-Brain" Axis-Related Mediators

Objectives The aim of this study is to investigate the effect of bisphenol A(BPA)and bisphenol B(BPB)exposure during pregnancy on placental-derived progesterone(Pg).And as important mediators of the "placenta-brain" axis,we also explore whether5-hydroxytryptamine(5-HT)and dopamine(DA)in placenta are affected by BPA and BPB,providing new evidence for the mechanism of neurotoxicity and developmental toxicity of bisphenol compounds.MethodA total of 212 pregnant women who gave birth in Wuhu Maternal and Child Health Care Hospital were enrolled in this study.The general information of pregnancy,serum,delivery data and full-term placenta were collected.The serum BPA and BPB exposure concentrations were detected by ultra-high performance liquid chromatography triple quadrupole mass spectrometry.According to its concentration,the subjects were divided into four groups: low exposure group(control group,n=54),combined high exposure group(double high group,n=55),high BPA exposure group(high BPA group,n=51),and high BPB exposure group(high BPB group,n=52).Placental Pg,5-HT and DA concentrations were measured by enzyme-linked immunosorbent assay.Analysis methods: According to the data type,chi-square test was used to compare the differences in gender distribution of infants born with different demographic characteristics,and the differences in the distribution of BPA and BPB during pregnancy with different demographic characteristics.Analysis of variance and t test were used to compare the differences in the distribution of Pg,5-HT and DA with different demographic characteristics.Chi-square test and analysis of variance were used to compare the differences in placental morphological parameters,delivery outcome,progesterone,and "placenta-brain" axis related mediators between different exposure groups.Linear regression model and Logistic regression model were used to analyze the association between BPA and(or)BPB exposure during pregnancy and placental morphological parameters,delivery outcome,progesterone,and "placental-brain" axis related mediators.ResultsThe results showed that there were no significant differences in the distribution of gender at birth and maternal age,BMI classification and other demographic characteristics(P > 0.05).There were no significant differences in the distribution of BPA and BPB exposure during pregnancy and maternal age,BMI classification and other demographic characteristics(P > 0.05).The risk of cesarean section was 2.05times(OR=2.05,95%CI: 1-4.21)higher in women with high BPA exposure during pregnancy than that in women with normal delivery.There were no significant differences in the distribution of Pg,5-HT and DA in other demographic characteristics(P > 0.05).BPA exposure during pregnancy was positively associated with placental5-HT level(β=0.1,95%CI: 0-0.19).Compared with the low BPB exposure group,the placental short diameter(β=-0.91,95%CI:-1.75-0.08)and Pg level(β=-0.25,95%CI:-0.46-0.04)in the high BPB exposure group were smaller.Compared with the pregnant women with low serum BPA and BPB levels during pregnancy,the high BPA,high BPB,and both high BPA and BPB groups had significantly lower birth weight,lower placental Pg levels,and higher placental DA levels(P < 0.05).ConclusionIn this study,high BPA exposure during pregnancy was positively correlated with 5-HT and DA levels and negatively correlated with Pg levels in full-term placental tissue.High BPB exposure during pregnancy was positively correlated with DA levels and negatively correlated with Pg levels in full-term placental tissue.No association was found between BPB and 5-HT.In addition,this study found that high exposure to BPA and/or BPB during pregnancy resulted in lower birth weight,and lower short diameter of full-term placenta in women with high exposure to BPB during pregnancy.This suggests that BPA and BPB may affect the neural development of offspring brain through the "placenta-brain axis" mediated by progesterone and neurotransmitters.
