| Objective : To study the expression level of micro RNA-1275 in Fadu cells,investigate the influence of mi R-1275 expression level in laryngeal carcinoma cells on tumor cell proliferation,migration and invasion,further clarify the role of micro RNA in the pathogenesis of laryngeal cancer,help to develop effective biomarkers and drug therapeutic targets for laryngeal cancer diagnosis,improve the clinical diagnosis and management strategy of laryngeal cancer,and improve the early diagnosis rate and overall survival rate of laryngeal cancer patients.Method : 1,Fadu cells were transiently transfected,and Import of the mi RNA-1275over-expressed vector(mimic)and its control sequence and inhibitory vector(inhibitor)and its control sequence into the target cells,The transfected cells were divided into four experimental cell groups,including Mi R-1275 mimic control group,Mi R-1275 inhibitor control group,Mi R-1275 mimic group and Mi R-1275 inhibitor group,Compared with blank control(blank)cells what was not transfected with any sequence.2,Detected the expression of mi R-1275 in five groups of cells by means of quantitative real-time PCR(Real-Time PCR,RT-PCR),;3,Routine culture of cells for 24hours and 48 hour after cell transfected,then conduct Cell Counting Kit-8(CCK-8).Calculate the cell viability of laryngeal carcinoma cells after the expression level of mi R-1275;4,after the transfected cells were routinely cultured for 24 h,evaluate the effect of mi R-1275 expression level in laryngeal carcinoma cancer cells by trans well cell invasion assay;5,measure the cell scratch healing rate of cells after 48 h of conventional culture in each group by cell scratch assay,in order to evaluate the effect of the expression level of mi R-1275 on the migratory ability of laryngeal carcinoma cells.Results : The over-expressed system and inhibitory expression system of mi R-1275 in laryngeal carcinoma Fadu cells were successfully constructed,and RT-PCR showed a significant decrease of mi R-1275 expression in Fadu cells.No significant differences in cell proliferation,invasion,and migration ability between mi R-1275 mimic control cells and mi R-1275 inhibitor control cells compared to the blank control group(P> 0.05).However,the proliferation,migration and invasion of cells in the mi R-1275 mimic group were significantly lower than those in the blank control group,while the proliferation,invasion and migration of cells in the mi R-1275 inhibitor group were significantly improved compared with the blank control group.Conclusion : Mi R-1275 is a key gene that regulates the activity of laryngeal cancer cells and plays an inhibitory role in the occurrence and development of laryngeal cancer.The further study of the expression level of mi R-1275 in laryngeal carcinoma tissue or serum and the targets in laryngeal carcinoma cell lines will promote great progress and break-through in the clinical diagnosis and treatment of significant cancer patients. |
