Expression And Significance Of EIF2B1 In HBV-associated Hepatocellular Carcinoma | | Posted on:2024-01-23 | Degree:Master | Type:Thesis | | Country:China | Candidate:W Y Qi | Full Text:PDF | | GTID:2544307082469384 | Subject:Internal Medicine (Infectious Diseases) | | Abstract/Summary: | PDF Full Text Request | | Objective Hepatocellular carcinoma(HCC)is one of the most common malignant tumors in human body.Among the causes of HCC,more than half of HCC cases are due to HBV infection,and liver cancer has attracted widespread attention due to its insidious onset,poor prognosis,and increasing global incidence.Protein translation plays an important role in the development of tumor.Research on molecular markers has become a hot topic at present.In this study,an karyotic translation initiation factor 2B1 is explored.The expression of eIF2B1 in HBV-related HCC and its clinical correlation provide reference for the early detection and clinical treatment of liver cancer.Methods Through the follow-up of clinical data of 743 patients with hepatocellular carcinoma in the specimen bank,91 patients with HBV related hepatocellular carcinoma with complete follow-up information were selected and their postoperative tumor tissues were selected.At the same time,paracarcinoma tissues larger than 2 cm away from the tumor margin were collected and prepared into 4 groups of tissue chips,and immunohistochemical staining was performed on 4 groups of tissue chips.At the same time,the expressions of eIF2α,P-eIF2α and eIF2B1 in Hep G2 and Hep G2.2.15 cells were detected by Western blot experiment.Then,Image J was used to analyze the proportion of positive cells stained by tissue chip and the gray value of Western blot results.Statistical analysis of clinical data and follow-up data of included patients was performed using Graphpad Prism8.0.2 and R4.0.5 software.Clinical data of included HBV-associated HCC patients were tested by nonparametric test to analyze the correlation between eIF2B1 expression and clinical characteristics in HBV-associated HCC patients.Survival curves were drawn to analyze the correlation between eIF2B1 expression and clinical prognosis in HBV-associated HCC patients.Multivariate regression analysis was conducted to establish a Cox proportional risk model,and Nomogram tandem table was constructed to evaluate the predictive value of eIF2B1 for the clinical prognosis of HBV-related HCC patients.Results By non-parametric test,the results showed that the expression of eIF2B1 in HCC tumor tissue had statistical significance with the number of HCC patients(P<0.05),but had no statistical significance with gender,age,maximum tumor diameter and stage(P>0.05).Immunohistochemical semi-quantitative results suggested that eIF2B1 was more strongly expressed in HCC tissues than in paracancer tissues.Western blot results showed that eIF2B1 was more strongly expressed in Hep G2.2.15 cells than Hep G2 cells.The difference of expression between the two groups was statistically significant(P<0.05).Survival analysis indicated that the expression of eIF2B1 in HBV-associated HCC had no significant statistical significance in overall survival of HCC patients(P>0.05),but in HBV-DNA positive patients,eIF2B1 expression was significantly correlated with overall clinical prognosis of HCC patients(P<0.05).The overall survival rate of the group with high eIF2B1 expression was lower than that of the group with low eIF2B1 expression.Cox multivariate regression analysis showed that eIF2B1 expression was an independent risk factor for overall clinical outcome in HBV-DNA positive patients(HR=4.28,P=0.018,<0.05).Conclusion The expression of eIF2B1 is significantly enhanced in HBV-associated HCC tissues,and is significantly correlated with overall survival in HBV DNA positive patients. | | Keywords/Search Tags: | Hepatocellular carcinoma, HBV, eIF2B1, eIF2 alpha, Protein translation | PDF Full Text Request | Related items |
