Objective:This study aims to analyze the expression of 9P21 gene and Ki67 protein in cervical lesions of different severity at different expression levels,and compare the diagnostic accuracy of single detection and combined detection,so as to achieve the purpose of accurate diagnosis and treatment of cervical lesions at a deeper level.Methods:Low-grade group:Tissue samples of patients who underwent cervical biopsy,cervical conization or hysterectomy due to cervical intraepithelial lesions in our hospital from September 2020 to February2022 were randomly selected,and postoperative pathological results returned as low-grade intraepithelial lesions in 37 cases.High-grade group:Tissue samples of patients who underwent cervical biopsy,cervical conization or hysterectomy due to cervical intraepithelial lesions in our hospital from September 2020 to February2022 were randomly selected,and postoperative pathological results returned as high-grade intraepithelial lesions in 56 cases.Control group(chronic cervicitis group):At the same time,28 patients who underwent total hysterectomy due to benign uterine lesions at the same time were randomly selected,and the postoperative pathological results returned as tissue samples of chronic cervicitis.9P21 gene was detected by FISH and Ki67 protein was detected by IHC method.The expressions of 9P21 gene and Ki67 protein were recorded in cervical lesions of different severity.P<0.05 was considered to be statistically significant.Results:1.There was no significant difference in age among all groups(F=0.093,P=0.911>0.05).There was no significant difference in the first sexual history(F=0.218,P=0.804>0.05).2.Age and first sexual history had no significant difference in the positive expression of 9P21 gene(χ2=4.012,χ2=0.168,P>0.05)and Ki67protein(χ2=0.505,χ2=0.418,P>0.05).3.The positive expressions of 9P21 gene and Ki67 protein were not equal among the three groups,and the positive expressions among the three groups were statistically significant(χ2=78.371,χ2=141.346,P <0.05).4.The positive expressions of 9P21 gene and Ki67 protein were positively correlated with the severity of cervical lesions at different levels(r=0.727,r=0.875,P<0.05),the positive expression rate of chronic cervicitis group was the lowest,and the positive expression rate of high-grade group was the highest.5.9P21 gene and Ki-67 protein had good diagnostic significance for HSIL;The 9P21 gene has good diagnostic significance for LSIL.The diagnostic significance of 9P21 in parallel with Ki67 was the greatest(AUC=0.852).The significance of 9P21 in tandem with Ki67 for HSIL diagnosis was greatest(AUC=0.928).Conclusions:1.The positive expression of 9P21 gene and Ki67 protein has clinical guiding value for the pathological grading diagnosis of HSIL to a certain extent.2.The positive expressions of 9P21 gene and Ki67 protein were positively correlated with the different severity of cervical lesions,which was helpful to predict the development rate and severity of cervical lesions and achieve more accurate diagnosis and treatment.3.9P21 gene combined with Ki67 protein contributes to the pathological grading diagnosis of cervical squamous intraepithelial lesions and reduces the rate of missed diagnosis and misdiagnosis.
