| Objective:Hepatic cellular cancer(HCC)is a serious threat to human health around the world.Its pathogenic factors are multiple and complex,including poor living habits such as smoking and excessive drinking,as well as factors such as diabetes and hepatitis.The infection of various hepatitis viruses is the main cause of the disease.viral hepatitis type B(HBV)has many transmission modes,strong infectious ability and high susceptibility.From the perspective of the whole world,more than 200 million people have been or are being infected by hepatitis B virus.Hepatitis B virus(HBV)infection can cause liver damage,which can progress to cirrhosis and eventually liver cancer.Liver cancer caused by hepatitis B virus(HBV)accounts for 50% of HCC in my country.Viral hepatitis type C(HCV)can now be cured in today’s medical environment.However,about half of infected patients will develop HCC.Although current treatments for HCC are continuously improving,the mortality rate of HCC remains high.Therefore,finding the pathogenesis and therapeutic targets of HCC has become a focus of research for experts and scholars.In many studies by experts and scholars,micro RNAs have been verified to exist as regulatory factors for cancer.Many studies have shown that micro RNAs can affect the occurrence and development of cancer.micro RNA-342-3p(miR-342-3p)has been shown in multiple studies to be associated with breast cancer,gastric cancer,and colon cancer,while micro RNA-2116-3p(miR-2116-3p)has been determined to have relevance to nasopharyngeal cancer and colon cancer in multiple articles.Although both of them act as tumor suppressors in cancers,there are few reports on the effects of these two micro RNAs on HCC.TTherefore,we detected the expression levels of miR-2116-3p and miR-342-3p in HCC tissues,analyzed their correlation with various clinical indicators,tumor differentiation degree and surgical prognosis of HCC patients,and discussed their clinical significance.Methods:In this paper,57 patients who underwent liver cancer resection in the Department of Intrahepatic Bile Duct of Chengde Medical College were selected as the research objects.Liver cancer and adjacent non-hepatic cancer tissues were collected and divided into two groups.There are 39 males and 18 females.This study intends to use q RT-PCR technology to analyze miR-342-3p and miR-2116-3p in liver cancer tissues and compare them.The sensitivity and specificity of these two miRNAs were analyzed.The experimental results were statistically analyzed in combination with clinical case data,with a significance level set at P<0.05.Results:1.The expression differences of miR-342-3p and miR-2116-3pThe expression of miR-2116-3p in HCC tissues was significantly lower than that in adjacent tissues(0.01 ± 0.02 vs 0.08 ± 0.26,P=0.034),and miR-342-3p(1.12±2.13 vs 7.97±11.32,P=0.034)was significantly lower than that in adjacent tissues.P<0.001)in HCC tissues were significantly lower than those in adjacent tissues.2.Diagnostic efficacy of miR-2116-3p and miR-342-3p in HCCThe area under the ROC curve(AUC)of miR-2116-3p and miR-342-3p for identifying HCC were 0.694(95% CI: 0.597-0.791,P=0.049)and 0.781(95% CI: 0.696-0.867,P=0.044),respectively.The calculated cutoff values were 0.85(sensitivity: 73.7%,specificity: 75.4%)and 0.0003(sensitivity:80.7%,specificity: 55.4%).The combined AUC of the ROC curve for identifying HCC from the control group was 0.793(95% CI: 0.711-0.879,P=0.042).3.The relationship between miR-2116-3p and miR-342-3p content in tissue and clinical pathological parameters of patientsUsing the median relative expression levels of miR-2116-3p and miR-342-3p in HCC tissues as the grouping basis,57 HCC patients were divided into two groups: low-expression group(n=29)and high-expression group(n=28).The clinical and pathological data of patients in different groups were compared.The results indicated that miR-2116-3p and miR-342-3p were significantly correlated with tumor size,TNM staging,and patient CLIP score.4.Kaplan-Meier curve was drawn to analyze the relationship between the expression of miR-2116-3P and miR-342-3p in tissues and poor prognosisThe incidence of poor prognosis in the miR-2116-3p low expression group was higher than that in the miR-2116-3p high expression group(P=0.021).There was no significant difference in the incidence of poor prognosis between the miR-342-3p low expression group and the miR-342-3p high expression group(P>0.05).5.Cox regression was used to analyze the risk factors for poor prognosis of HCCUnivariate Cox regression analysis showed that the presence or absence of tumor thrombus,CLIP score,and miR-2116-3p expression were all risk factors for poor prognosis of HCC patients.Multivariate Cox regression analysis showed that the presence or absence of tumor thrombus and the expression of miR-2116-3p could be used as independent risk factors for predicting poor prognosis of HCC patients.Conclusions:miR-2116-3p and miR-342-3p are potential diagnostic markers for HCC,which are beneficial supplement to the traditional marker AFP.The expression levels of miR-2116-3p and miR-342-3p are significantly correlated with tumor size,TNM stage and CLIP score.The expression level of miR-2116-3p can predict the poor prognosis of patients,which is an independent risk factor for the poor prognosis of patients. |
