Objective: The purpose of this study is to collect and summarize Professor Cao Enze’s medication rules,drug association degree and cluster analysis in the treatment of uric acid nephropathy(UAN)through the auxiliary platform of traditional Chinese medicine inheritance,mine the drugs based on Qingbu Tongluo method to treat UAN,and obtain the core prescription-Qingbu Tongluo Formula.Finally,by means of network pharmacology,the mechanism of Qingbu Tongluo Formula in the treatment of UAN is expounded,providing a reliable theoretical basis for the effective treatment of uric acid nephropathy with Qingbu Tongluo Formula.Method:(1)Collect the medical record data of patients who met the inclusion criteria of this study in the outpatient service of Professor Cao Enze from October 2020 to December2022,standardize the diagnosis name and drug name,and then enter the data into the auxiliary platform of traditional Chinese medicine inheritance,analyze the frequency of drug use,drug taste,meridian tropism and efficacy,complete the drug association analysis and cluster analysis and complex network diagram processing,and finally get the Qingbu Tongluo fang as the core group.(2)After integrating the database information,a total of 107 active ingredients and 386 drug targets were selected from Qingbu Tongluo Formula,and 1561 disease target genes were obtained.After the intersection,138 drug-uric acid nephropathy intersection targets were obtained,and the core components were quercetin β-Sitosterol,stigmasterol,luteolin,kaempferol.Through the PPI protein target map,the key targets are TNF,IL6,IL1 B,TP53,VEGFA,PPARG,CASP3,PTGS2,MMP9,CXCL8,CCL2,H1F1 A.Through the GO enrichment bar chart,it can be found that the intersection target of Qingbu Tongluo Formula in treating uric acid nephropathy can positively regulate cell apoptosis,regulate the expression of lipopolysaccharide hypoxia cell response gene,and regulate the transcription of RNA polymerase II promoter.KEGG enrichment analysis showed that the main mechanism of Qingbu Tongluo Formula in treating uric acid nephropathy was quercetin β-Glutosterol,luteolin and other components act on many important targets such as PTGS2,PIK3 CG,IL1B,TNF,BCL2,RELA,CASP3 and so on through PI3K-Akt,AGE-RAGE,lipid and atherosclerosis,MAPK,TNF,NF kappa B and other related signal pathways to achieve therapeutic effects.Result:(1)A total of 192 patients were included,including 115 males and 77 females.The ratio of men and women is 1:1.493;The age group is mainly distributed in 41-60 years old.According to statistics,there are 103 kinds of traditional Chinese medicine for treating uric acid nephropathy,and the traditional Chinese medicine used more than 100 times includes: astragalus,scorpion,cicada slough,rhubarb,earthworm,atractylodes atractylodes,zombie silkworm,calcined keel,poria cocos,oyster,amomum villosum,panax notoginseng,bergamot,and corn whisker.The drug properties are mainly cold,warm and mild.The medicine mainly tastes bitter,sweet and spicy.The meridians of traditional Chinese medicine with a frequency of>10% mainly include liver,spleen,stomach,lung and kidney meridians.The most frequently used drugs are the drugs for calming the liver and calming the wind.The representative drugs include scorpion,earthworm,zombie silkworm and oyster.Through complex network analysis,a core ingredient(13 flavors in total)was obtained and named Qingbu Tongluo Formula.(2)After integrating the database information,a total of 107 syndrome of deficiency and excess.Professor Cao Enze takes "clearing and tonifying the collaterals" as the basic treatment of this disease,which is also the origin of the clearing and tonifying the collaterals.(2)Through data mining,the core group of 13 Chinese herbs in the whole prescription are displayed as follows: astragalus,amomum villosum,corn whisker,zombie silkworm,cicada slough,herba siegesbeckiae,scorpion,atractylodes,rhubarb,earthworm,tuckahoe,panax notoginseng and zedoary rhizome.(3)The network pharmacological research further verified that the basic components of Qingbu Tongluo Formula in treating uric acid nephropathy are quercetin β-Sitosterol,stigmasterol,luteolin,kaempferol;The key targets are TNF,IL6,IL1 B,TP53,VEGFA,PPARG,CASP3,PTGS2,MMP9,CXCL8,CCL2,H1F1 A.Through GO functional annotation,it can be found that the intersection target of Qingbu Tongluo Formula in treating uric acid nephropathy can positively regulate cell apoptosis,regulate gene expression of lipopolysaccharide hypoxia cell response,and regulate transcription of RNA polymerase II promoter.KEGG enrichment analysis showed that the main signal pathways of Qingbu Tongluo Recipe in treating uric acid nephropathy were PI3 K Akt,AGE-RAGE,lipids and atherosclerosis,MAPK,TNF,NF kappa B,etc.
