Objective: To preliminarily explore the relationship between urinary protein and stones by analyzing the stone matrix and urinary protein of oxalate stone patients,and examining the concentration of S100A8/A9 in the urine of stone patients and healthy people.Methods: Protein data of calcium oxalate(Ca Ox)stones and urine of calcium oxalate stone patients were collected from proteomics databases,and the latest newly released Uniprot protein database was used to identify the collected stone proteomics data.The proteins with stable content in the stone matrix proteins(SMP)were screened out.20 proteins were further analyzed using random forest,protein interaction NETswork,GO function and KEGG pathway enrichment.The results were comprehensively analyzed to find S100 Calcium Binding Protein A8/A9(S100A8/A9)protein that has a greater impact on stones.Stones and urine from stone patients treated by urology department of Affiliated Hospital of Youjiang Medical University for Nationalities were collected,and infrared spectrometer was used to identify the nature of stones to exclude non-calcium oxalate stones.Urine samples from healthy people who came to the hospital for physical examination during the same period were collected,and enzyme-linked immunosorbent assay(ELISA)method was used to measure S100A8/A9 content in urine of calcium oxalate stone patients and healthy people.SPSS software was used for statistical analysis,and two independent sample t-test was used to compare S100A8/A9 concentration difference between two groups of urine.Results:(1)By analyzing the proteomics of Ca Ox stones,a total of 20 stable and commonly present proteins were identified in the stones.(2)GO and KEGG enrichment analysis showed that these 20 common proteins were mainly associated with inflammatory response,ion binding,antioxidant activity,and other related functions.(3)Random forest analysis identified the top 5 important proteins,which were ranked as S100A9,S100A8,ALB,KRT9,and IGHAl.(4)Measurement of S100A8/A9 concentrations in urine samples showed that the levels of S100A8/A9 were significantly higher in the urine of Ca Ox stone patients than in healthy controls(P < 0.003).Conclusion:(1)The proteins with stable content in Ca Ox stones are: HBB,DEFA3,KRT9,AZU1,RNASE3,CTSG,ELANE,S100A9,MPO,S100A8,KRT1,ALB,APCS,IGHM,IGKC,C3,IGHG2,IGHA1,IGHG3,SERPINA1.(2)S100A8/A9 protein exists in the urine of both the Ca Ox stone disease group and the healthy control group.The S100A8/A9 content in the urine of the Ca Ox stone group is higher than that of the healthy control group,and the results are statistically significant. |
