Objective: To investigate the effect of matrine on invasion and migration of SW480 colon cancer cells and the expression of TGF-β1 and Smad related proteins.Methods:(1)In vitro culture SW480 colon cancer cells,adding matrine in different concentrations(0 mg/m L,0.25 mg/m L,0.5 mg/m L,0.75 mg/m L,1 mg/m L,1.25 mg/m L,1.5mg/m L),The effect of matrine on SW480 colon cancer cell viability was determined by CCK-8 assay.(2)The inhibitory effect of matrine(0 mg/m L,0.5 mg/m L,0.75 mg/m L,1mg/m L)on the lateral migration of cells was determined by cell scratch method.(3)Transwell migration assay was used to measure the longitudinal migration ability of cells,and invasion assay was used to measure the invasion ability of cells.(4)The protein expression levels of E-cadherin,N-cadherin,TGF-β1,Smad2/3 and p-Smad2/3 were determined by Western blot.(5)SPSS 24.0 software and Graph Pad Prism8 were used for statistical analysis.All measurement data were expressed as mean ± standard deviation((?)±s),and differences between groups were analyzed by one-way ANOVA.P<0.05 was considered statistically significant.Results:(1)Compared with the control group,the cell viability of SW480 cells was significantly inhibited by different concentrations of matrine(0 mg/m L,0.25 mg/m L,0.5mg/m L,0.75 mg/m L,1 mg/m L,1.25 mg/m L,1.5 mg/m L).The results were concentration-dependent and time-dependent(P<0.05).(2)Matrine concentrations were 0mg/m L,0.5 mg/m L,0.75 mg/m L and 1 mg/m L in subsequent experiments.The cell scratch method showed that the lateral migration ability of cells decreased significantly with the increase of matrine concentration 48 h after treatment(P<0.05).(3)Transwell migration and invasion assay showed that invasion ability and longitudinal migration ability of cells were significantly decreased compared with control group(P<0.05).(4)Western blot results showed that after treatment with different concentrations of matrine for 48 h,compared with the control group,EMT related proteins were: The expression level of E-cadherin protein was increased,the expression level of N-cadherin protein was decreased(P<0.05),and the expression levels of TGF-β1,Smad2/3,P-Smad2/3 protein were decreased(P<0.05).Conclusion: Matrine inhibited the proliferation of SW480 colon cancer cells in a time-and concentration-dependent manner.Matrine can inhibit the invasion and migration of SW480 colon cancer cells,increase the expression level of E-cadherin protein,decrease the expression level of N-cadherin protein,and inhibit the expression of TGF-β1,Smad2/3,p-Smad2/3 protein.These results suggest that the inhibitory effect of matrine on SW480 colon cancer cells may be related to the influence of TGF-β1/Smad signaling pathway.
