Objective:To study the angiogenesis in the operative area of Oral leukoplakia with Oral submucous fibrosis(OSF)treated by Photodynamic therapy(PDT),and to explore the prospect of the application of angiogenesis after PDT in the treatment of OSF.Methods: The OSF-affected lesions were randomly collected from20 patients with both OSF and OLK before and after PDT treatment.The expression of Vascular endothelial growth factor(VEGF)in OSF lesions before and after PDT was evaluated by immunohistochemical staining;Using vascular endothelial marker CD34 to detect the mean Microvessel density(MVD),mean Microvessel area(MVA)and mean microvessel perimeter(MVLP)of positive microvessels in OSF lesions before and after PDT operation.At the same time,the mean Microvessel density of labeled microvessels in OSF lesions before and after PDT treatment was detected by vascular endothelial marker CD105,and the collagen fibers in submucosal lamina propria in OSF lesions were automatically analyzed by computer-aided technology after Masson special staining,and the Mean optical density(MOD)of submucosal lamina propria was measured before and after PDT treatment.Results: The final scores of VEGF expression in lesion tissue of patients before and after PDT treatment was 6.851±2.534 and9.997±1.869,respectively.The expression of VEGF was significantly higher after PDT than before operation(P=0.0002).For the microvessels labeled by CD34,the CD34-MVD in the affected tissue of the patients before and after PDT was 19.633±20.469 and 69.583±17.141,respectively,and the value of CD34-MVD after PDT was significantly higher than that before PDT(P=0.004);and the CD34-MVA of the affected tissue before and after PDT was 0.000124±0.000051 and0.000171±0.000042,respectively,the value of CD34-MVA after PDT was slightly higher than that before operation(P=0.0074);The CD34-MVLP before and after PDT was 0.041±0.016 and 0.050±0.011 respectively,but the difference was not statistically significant(P=0.0586).For the microvessels labeled by CD105,the CD105-MVD of the affected tissue samples before and after PDT was 9.750±5.692 and25.150±6.655 respectively,the value of CD105-MVD after PDT was significantly higher than that before PDT(P=0.021).After Masson special staining,the MOD was analyzed and calculated by computer software.the value of MOD submucous lamina propria of affected tissue samples before and after PDT were 0.09154±0.02040 and 0.07423±0.02951,respectively,and the MOD values after PDT were significantly lower than those before PDT(P=0.000).Conclusion: Photodynamic therapy can promote the angiogenesis of OSF lesion mucosa,which provides a theoretical basis for photodynamic therapy for Oral submucous fibrosis.
