| Objective:(1)To explore the diagnostic efficacy of liquid biopsy miRNA in endometrial cancer by meta-analysis。(2)To explore the role and possible target of miR-182 in endometrial cancer。Method:(1)Using R and revman5.3 software for meta-analysis of the sensitivity,specificity and the area under the curve(AUC)of miRNA in the diagnosis of endometrial cancer。(2)Using Kaplan Meier plotter database and starbase3.0 database analysis of multiple miRNAs to evaluate the prognosis of endometrial cancer.(3)miRDB、Target Scan、GEPIA online tools were used to screen the key target genes of mir-182 in endometrial cancer,and R as used for enrichment analysis。(4)q RT-PCR was used to detect the expression levels of FAM171A2、ATP6V1F、RPUSD1 in Ishikawa Cells,endometrial cancer tissue and normal endometrium。(5)The possible binding sites of miR-182 were screened by miRBase and miRDB database。Result:(1)The sensitivity and specificity of miRNA in the diagnosis of endometrial cancer were 0.79(95% CI: 0.75-0.83)and 0.82(95% CI:0.76-0.86);The area under the comprehensive curve is 0.8,which has higher diagnostic efficiency 。 Combined miRNA has higher diagnostic efficiency in the diagnosis of endometrial cancer than single miRNA。(2)The survival rate of endometrial cancer patients with high expression of miR-182 was higher than that of patients with low expression of miR-182(P < 0.05)。(3)The expressions of FAM171A2,ATP6V1 F and RPUSD1 in endometrial cancer and Ishikawa Cells were higher than those in normal endometrium(P < 0.01)。(4)RPUSD1 has the binding site of miR-182 and has high binding efficiency。Conclusion:(1)The blood miRNA of liquid biopsy is relatively stable and easy to collect。It has high diagnostic efficiency in endometrial cancer.It is a diagnostic biomarker with high sensitivity and specificity。(2)miR-182 plays a role in endometrial cancer,and there is a targeted relationship between miR-182 and RPUSD1。... |
