Object:To profile the role of circ RNA in regulating peripheral T cells and investigate the underlying mechanism in Immune Thrombocytopenia(ITP).Methods:Peripheral blood of ITP patients and healthy controls were collected to isolate the peripheral T cells.High throughput sequencing was used to profile the circular RNA in peripheral T cells.we conducted enrichment analysis for differential circ RNA using Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG).A spectrum of target prediction websites were used to analyze the downstream targets of circular RNA or mi RNA with dual luciferase reporter assay for validation.Lentivirus transfection was conducted for overexpression or knockdown of mir-641.In vivo,the level of mir-641 in ITP model mice was restrained using tail vein injection of antagomir-641.The proportion of Th17 and Treg in CD4~+T cells was detected by flow cytometry.q PCR and Western Blotting experiments were utilized to analyze m RNA and protein expression.Conclusion:A unique circular RNA profile was found in peripheral T cells of ITP.Circular RNA is closely related to metabolism and receptor activation of peripheral T cells in ITP.Circular RNAs may be the potential biomarkers and targets for the diagnosis and treatment of ITP.hsa_circ_0008866 may play a role in the imbalance of the Th17/Treg via the mi R-641-SATB1 axis.Targeting hsa_circ_0008866—mi R641—SATB1axis may be a new strategy for the therapy of ITP. |
