Purpose: To investigate the effects of cilengitide on the expression level of high mobility group protein B1(HMGB1)in LPS and ATP-induced mouse macrophage RAW264.7 cells in vitro.Methods: Mouse macrophage RAW264.7 cells were divided into three group: control group,inflammation model group(LPS+ATP),and cilengitide intervention group(LPS+ATP+cilengitide).The m RNA expression levels of HMGB1 and IL-1β were tested by RT-q PCR,and the protein expression levels of HMGB1 were messured by Western Blot,and the concentrations of supernatant proinflammatory cytokines IL-1β,IL-6and TNF-α were determined by ELISA.Results:(1)After mouse macrophage RAW264.7 cells were stimulated by LPS and ATP,the m RNA expression levels of IL-1β and HMGB1,the protein expression levels of HMGB1 and the concentrations of supernatant proinflammatory cytokines IL-1β,IL-6 and TNF-α in inflammatory model group were significantly higher than those in control group(P all<0.0001).(2)Compared with these in the inflammatory model group,the m RNA and the protein expression levels of HMGB1 in cilengitide intervention group were significantly decreased(P both<0.0001),but the m RNA expression levels of IL-1β were not significantly different;the concentrations of supernatant proinflammatory cytokines IL-1β,IL-6 and TNF-α in cilengitide intervention group were significantly decreased(P all < 0.0001).Conclusions:Cilengitide could inhibite the expression of HMGB1 and the release of proinflammatory cytokines IL-1β,IL-6 and TNF-α,thus alleviating the inflammatory response of mouse macrophages.
