Objective : The incidence and mortality of bladder cancer are increasing year by year,seriously endangering human health.Many studies have shown that lnc RNAs are closely related to occurrence and development of cancers.Lnc RNA LINC00467 can promote the occurrence and development of various malignant tumors,but the role of LINC00467 in bladder cancer has not yet been elucidated.This study aims to explore the expression,biological function and mechanism of LINC00467 in bladder cancer,and to explore new diagnostic markers and therapeutic targets for bladder cancer.Methods:1.We explored the expression of LINC00467 in bladder cancer tissue by analyzing TCGA and GEO databases,detected the expression level of LINC00467 in bladder cancer and adjacent normal tissues by q RT-PCR,and analyzed the expression level of LINC00467 in bladder cancer patients with GEPIA database.Correlation.2.Taking the bladder cancer RT4 and T24 cell lines as the research objects,the clone formation assay,MTT assay,scratch assay and Transwell assay were performed to explore the effect of LINC00467 on the proliferation,migration,invasion and malignant phenotype of bladder cancer.3.Using bioinformatics analysis,fluorescence in situ hybridization(FISH),RNA pull-down,RNA Binding Protein Immunoprecipitation(RIP),Western blotting and other experiments to explore the regulation of LINC00467 Specific mechanisms of the malignant phenotype of bladder cancer.4.Functional recovery experiments confirmed that LINC00467 promotes bladder cancer proliferation,migration and invasion by regulating NF-κb signaling pathway.5.The function of LINC00467 in promoting the proliferation of bladder cancer was confirmed by subcutaneous xenograft tumor model in nude mice.Results:1.Compared with adjacent normal tissues,the expression of LINC00467 was significantly increased in bladder cancer tissues,and was negatively correlated with the prognosis of patients.2.Phenotyping experiments showed that after knockdown of LINC00467,the proliferation,migration and invasion of RT4 and T24 cells were significantly decreased;while overexpression of LINC00467,the proliferation,migration and invasion of cells were significantly increased.3.LINC00467 can increase its stability by binding to NF-κb-p65 m RNA;and promote the activation and translocation of NF-κb-p65 protein by binding to it,while enhancing its stability.4.Functional recovery experiments showed that LINC00467 could promote the migration and invasion of bladder cancer by regulating the NF-κb signaling pathway.5.In the nude mouse subcutaneous xenograft model,the volume and weight of the xenografted tumor in the overexpression LINC00467 group were significantly larger than those in the control group,while those in the knockdown LINC00467 group were significantly smaller than those in the control group.Conclusions : LINC00467 is highly expressed in bladder cancer tissues and is negatively correlated with the prognosis of patients.LINC00467 can promote NF-κb-p65 protein activation and nuclear translocation to promote bladder cancer proliferation,migration and invasion.9charts,52references...
