Objective: To investigate B1 and Tfh cell levels in children with systemic lupus erythematosus(SLE)and whether mesenchymal stem cells(MSCs)can play a therapeutic role by regulating B1 and Tfh cells in the peripheral blood of children with SLE.Methods:(1)Peripheral blood mononuclear cell(PBMC)were isolated from human peripheral blood by high-speed centrifuge,and B1 cells and Tfh cells in PBMC were detected by flow cytometry analysis,and the cell differences between SLE patients and controls were analyzed,as well as the correlation between cell populations and clinically relevant indexes in SLE patients.(2)Use the MSCs dedicated serum-free medium for culturing human umbilical cord mesenchymal stem cells(h UCMSCs),and co-cultured MSCs with obtained PBMC according to PBS control group,h UCMSCs: PBMC(1:10)group and h UCMSCs: PBMC(1:20)group for 48 h.The ratio of B1 cells and Tfh cells after culture was detected again.Results:(1)hUCMSCs grow adherently and are capable of osteogenic and adipogenic differentiation.(2)B1 cells in SLE patients,higher than controls(P<0.05),did not correlate significantly with serum C3,serum C4,Ig G,Ig E,IgA,IgM,antinuclear antibodies,anti-double-stranded DNA antibodies,and SLEDAI scores(P>0.05).Tfh cells were higher in SLE patients than in controls(P<0.05),with no significant correlation with serum C3,Ig G,Ig E,Ig A,antinuclear antibodies,anti-double-stranded DNA antibodies,and SLEDAI scores,but a trend towards correlation with serum C4 and Ig M.(3)After co-cultured PBMC with h UCMSCs,there was no difference in B1 cells and Tfh cells between the groups(P > 0.05).Conclusions: The number of B1 cells and Tfh cells differed between the SLE group and the control group and may be involved in the development of SLE.h UCMSCs may not exert a therapeutic effect on SLE by regulating the number of B1 cells and Tfh cells in peripheral blood.
