Objective:To explore the expression and clinical significance of KIAA1429 in colorectal cancer tissues and cell lines,and to study the effect of KIAA1429 on the proliferation and migration of colorectal cancer cells in vitro.Methods:TCGA and GEPIA databases were used to analyze the expression of KIAA1429 in colorectal cancer and its relationship with clinicopathological features and prognosis.Immunohistochemistry were used to analyze the expression of KIAA1429 in colorectal cancer and its relationship with clinicopathological features.The expression of KIAA1429 in colorectal cancer was analyzed by Western blot.Univariate and multivariate analysis were used to evaluate the relationship between KIAA1429 expression and prognosis of CRC patients.The expression of KIAA1429 in colorectal cancer cell line SW480 was knocked down,and the transfection efficiency was detected by Western blot and q RT-PCR.The proliferation and migration of colorectal cancer cells were detected by CCK-8 method and cell scratch test.Results:The results of TCGA database analysis showed that amplification rate of KIAA1429 in colorectal cancer is about 6%,and ratio of colorectal cancer patients with KIAA1429 genetic alterations could reach above 10%.The results of TCGA and GEPIA database analysis showed that KIAA1429 was highly expressed in colorectal cancer tissues(P <0.05),and its high expression was related to TNM stage(P < 0.001)and Overall survival(P = 0.027).The results of immunohistochemistry and Western blot showed that KIAA1429 was highly expressed in colorectal cancer tissues,and its expression was significantly correlated with the lymph node metastasis(P <0.001)and tumor TNM stage(P=0.011).Multivariate Cox regression analysis showed that nerve invasion(P=0.001)and KIAA1429 expression level(P = 0.004)were related to the prognosis of patients.KIAA1429 is an independent prognostic factor in CRC patients.Knocking down the expression of KIAA1429 can significantly inhibit the proliferation and migration of CRC cell SW480 in vitro.Conclusion:The expression level of KIAA1429 in colorectal cancer tissue was significantly increased,and its high expression was significantly correlated with lymph node metastasis and tumor TNM stage.The overall survival of patients with KIAA1429 high expression colorectal cancer is shorter than that of patients with low expression.Inhibiting the expression of KIAA1429 can reduce the proliferation and migration of colon cancer cell SW480.
