Objective: Endometrial cancer(EC)is one of the most common tumors in women.EC patients with local or distant metastasis have a sharp decline in the 5-year survival rate.It may improve the diagnosis rate of patients,carry out risk stratification and prognosis judgment.This study aimed to analyze the relationship between the expression of PRDX4 and GRP78 and the clinical characteristics and prognosis of EC patients.Methods: The GEPIA online bioinformatics analysis tool was used to analyze the expression of PRDX4 and GRP78 in endometrial cancer patients in TCGA and GTEx databases and their relationship with patient prognosis.The cancer tissues and paracancerous tissue sections of 76 patients with endometrial cancer and 41 fresh EC tissues and their paracancerous tissues were collected,and the expressions of PRDX4 and GRP78 were detected by immunohistochemistry and real-time quantitative PCR,respectively.HE staining method to check the morphological characteristics of EC tissue and normal tissue.The chi-square test was used to analyze the relationship between PRDX4 and GRP78 and the clinical characteristics of patients,the Kaplan-Meier method was used to analyze the relationship between PRDX4 and GRP78 and the survival time of endometrial cancer patients,and the COX regression method was used to analyze the independent risk factors for the prognosis of endometrial cancer patients.Results: The expression levels of PRDX4 and GRP78 in endometrial cancer tissues were significantly increased(P<0.05),and the two were positively correlated(P<0.05).The expressions of PRDX4 and GRP78 in the endometrial cancer group were related to FIGO stage and distant metastasis(P<0.05);the co-expression of PRDX4 and GRP78 was an independent risk factor affecting the prognosis of endometrial cancer,and those with high PRDX4 and GRP78 expression.The overall survival time was significantly lower than those with low expression(P<0.05).Conclusion: The high expression of PRDX4 and GRP78 is associated with poor prognosis and clinicopathological characteristics of endometrial cancer patients,and there may be a synergistic effect between the two.
