Endophytic fungi can produce the same or similar active substances from host plants due to their long-term close symbiotic relationship with host plants,which is an important source of innovative drug discovery.Four strains with good antibacterial and antitumor activities were isolated from Pratia nummularia,Anredera cordifolia,Anaphalis sinica and Selaginella delicatula,which were Fusarium proliferatum TU-2110,Fusarium graminearum TSQ-2,Metapochonia bulbillosa XQ-1 and Shiraia sp.BYJB-1 respectively.In this paper,the above four endophytic fungi were taken as the research object,and their secondary metabolites were systematically studied in order to obtain natural active molecular compounds with novel structure and significant activity.Based on silica gel column chromatography,ODS,Sephadex LH-20column chromatography,prep-HPLC and other modern chromatographic separation methods,40 compounds were isolated and characterized by 1D/2D NMR,HRMS,CD/ECD and chemical derivatization,including 4 new compounds.The structural types include cyclic peptides,alkaloids,polyketones,terpenoids,steroids,etc.Eight monomer compounds(1-8)were isolated and identified from F.proliferatum TU-2110,including a new cyclic peptide(1)and a new indole natural product(3).Nine monomer compounds(9-17)were isolated and identified from F.graminearum TSQ-2.Thirteen monomer compounds(18-30)were isolated and identified from M.Bulbillosa XQ-1,including a new pyridone(18).Ten monomer compounds(31-40)were isolated and identified from Shiraia sp.BYJB-1,including a new benzophenone(31).The cytotoxicity and antibacterial activity of some isolated monomeric compounds were evaluated by the MTT method and 96 well broth dilution method respectively.The results showed that compounds 2,13,38 and 39showed different degrees of inhibitory activity on human Hep-G2 and SMMC-7721 hepatoma cell lines,with IC50 values ranging from 8.04 to 50.48μM.Compound 2 showed significant inhibitory effect on Staphylococcus aureus and Enterococcus faecalis,and the MIC values reached 7.8 and 15.6μg/m L,respectively.Finally,this paper reviews the structure and bioactivity of new compounds found in Fusarium fungi from 2015 to 2022,including 211 chemical components,including alkaloids,polyketones,terpenes and other structural types,and discusses the related activities.Figures 81,Tables 50,References 79...
