| Objective: To investigate the effect of micro RNA-93-5p(miR-93-5p)on retinal neuron apoptosis in acute ocular hypertension(AOH)by regulating PDCD4 gene.Methods: 1)Acute ocular hypertension rat model and oxygen and glucose deprivation / recovery(OGD/R)model of R28 cells were used for experimental exploration.2)TUNEL staining was used to evaluate the apoptosis of retinal neurons and R28 cells from the perspective of morphology.3)RT-q PCR(q PCR)was used to detect the expression of miR-93-5p and PDCD4 genes in AOH retina.4)Western blot(WB)was used to detect the expression of Bax,Bcl-2,PDCD4,PI3 K,Akt and p-Akt proteins in AOH retina.Results: Compared with the control group,the expression of miR-93-5p decreased and the expression of PDCD4 m RNA increased in the retina of AOH.And overexpression of miR-93-5p or interference with PDCD4 expression can significantly reduce the number of TUNEL positive R28 cells,which suggested that miR-93-5p and PDCD4 can regulate the apoptosis of retinal cells.Further experiments in vivo found that overexpression of miR-93-5p could decrease the ratio of Bax/Bcl-2and reduce the number of TUNEL staining positive cells in retinal neurons of rats with acute ocular hypertension.In addition,the m RNA and protein expression of PDCD4 in retina decreased after miR-93-5p was overexpress.Interfering with the expression of PDCD4 decreased the ratio of Bax/Bcl-2 and reduced the number of TUNEL staining positive retinal neurons.Overexpression of miR-93-5p or interference with PDCD4 expression could increase the expression of PI3 K,Akt and p-Akt pathway proteins.Conclusions: Under the condition of AOH injury,miR-93-5p reduced the apoptosis of retinal neurons by inhibiting the expression of PDCD4 further upregulating PI3K/Akt pathway.These results suggested that miR-93-5p and PDCD4 may become new potential targets for glaucoma treatment,which may provide new ideas for the treatment of glaucoma. |
