Objective:The aim of this study is to detect the expression level of BRD4 in Epithelial ovarian cancer(EOC)tissues and explore its relationship with clinicopathological characteristics,so as to provide a new theoretical basis for the clinical diagnosis and treatment of EOC.Methods:1.A total of 106 cases of ovarian specimens and clinical data(52 cases of EOC tissue,19 cases of benign ovarian tumor epithelial tissue,35 cases of normal ovarian epithelial tissue)were collected from a tertiary hospital of Jilin University from December 2021 to December 2022.All of them were diagnosed by professional pathologists after surgery.Surgical and pathological staging was performed by the International Federation of Gynecology and Obstetrics(FIGO).The patients were divided into malignant,benign and normal groups.Previous history of malignant tumor,use of steroid hormones in the last 3 months,incomplete clinical data and invalid tissue samples during staining were excluded from the three groups.In the malignant group,neoadjuvant therapy before surgery and metastatic malignant tumor of ovary after surgery were further excluded.In the end,31 cases of qualified EOC tissue were retained,12 cases of benign ovarian tumor epithelial tissue,and 28 cases of normal ovarian epithelial tissue were removed due to benign gynecological diseases.2.Immunohistochemistry was used to detect the expression of BRD4 in the three groups of specimens.Because BRD4 was mainly expressed in the nucleus,it was considered as positive expression when there were yellow,brown and tan particles in the nucleus,and negative expression when the cells were not stained or the cytoplasm was stained but the nucleus was not stained.Immunohistochemical image acquisition system was used to take pictures of the well-stained areas of the three groups of slides and store them in the computer system.Each slice will be enlarged by 400 x,and multiple pictures will be taken at random in different fields of view.Immunohistochemical staining in the three groups of specimens was subsequently quantified using Image analysis software Image j 1.8.After uploading the image to the software,the software can directly measure the Integrated optical density(IOD)and area of each image,and calculate the Average Optical Density(AOD).The ratio of the cumulative optical density value to the Area of the region,that is,AOD=IOD/Area.Three or more pictures were analyzed for each section,and the AOD was averaged for later analysis and discussion.3.SPSS 27.0 software was used for correlation analysis,and Kruskal Wallis H test was used for statistical analysis of BRD4 expression among the three groups.If the clinical and pathological features of the malignant group were in accordance with the normal distribution,the correlation between the clinicopathological features and BRD4 expression was analyzed by independent sample t test,and if the distribution did not conform to the normal distribution,the Mann-Whitney U test and Pearson correlation analysis of the non-parametric rank sum test were used.The analysis results were plotted using Graph Pad prism 9.0 mapping software.Results:1.Compared with normal ovarian epithelial tissues and benign ovarian tumor epithelial tissues,BRD4 is highly expressed in EOC tissues(P<0.05),and the difference was statistically significant.2.BRD4 protein expression is associated with FIGO stage in EOC patients.Compared with early-stage(I-II)ovarian cancer patients,BRD4 expression was significantly increased in advanced(III-IV)ovarian cancer tissues(P<0.05),and the difference was statistically significant.3.There was no significant difference in the expression level of BRD4 in EOC with serous carcinoma,lesion diameter,lymph node metastasis,ascites,distant metastasis,P53 mutation and Ki67 positive rate(P>0.05).4.The expression level of BRD4 in EOC was not correlated with the expression of CA125,CA199,HE4 and CEA(P>0.05).5.The expression level of BRD4 in EOC was not significantly correlated with age,BMI,parity and menopause(P>0.05).Conclusions:1.The expression level of BRD4 in EOC tissues is higher than that in normal ovarian epithelial tissues and benign ovarian tumor epithelial tissues,which is closely related to the occurrence and development of EOC.2.The expression of BRD4 is related to the FIGO stage of EOC,promotes the occurrence and development of epithelial ovarian cancer,can be used as an effective biomarker to evaluate the severity of the disease.3.BRD4 provides a new clinical basis for molecular targeted therapy of EOC.
