| Objective:The aim of this study is to investigate the alleviating effect of Lactiplantibacillus plantarum P101(P101)on lead-induced myocardial injury and to explore its underlying mechanism.Method:Mice,six weeks old and of the male C57BL/6 strain,were randomly distributed into three separate groups.including the normal diet group(ND),ND+Pb group,and ND+Pb+P101 group.Mice in the ND+Pb group and ND+Pb+P101 group were fed water containing Pb,while mice in the ND+Pb+P101 group were gavaged with 200μL bacterial suspension(2×10~8CFU of P101)to prevent cardiac lesions.The experiment lasted for 8 weeks,after which mice were euthanized by cervical dislocation and heart tissues were collected.By detecting serum biomarkers(ALAD,Tn-T,AST,ALT,TG,and CHO),myocardial pro-inflammatory cytokines(p-NF-κB,TNF-α,IL-1β,and PAI-1),myocardial apoptosis markers(caspase-3),myocardial injury markers(p21 and BNP),TH1/TH2 balance-related proteins(T-bet/GATA-3),and cardiac fibrosis-related indices(Sirius Red staining and collagen 1A1 expression),we aim to evaluate myocardial injury.Result:1.Compared to the ND group,the ALAD level was significantly reduced(P<0.001)and Tn-T,AST,and ALT values were significantly increased(P<0.05)in the ND+Pb group.In contrast,the ALAD level was significantly elevated(P<0.01)and Tn-T,AST,and ALT values were significantly reduced(P<0.05)in the ND+Pb+P101 group compared to the ND+Pb group.The changes in TG and CHO among all groups showed no statistically significant differences(P>0.05).2.Using Western blot technology,we investigated the expression levels of proteins such as p-NF-κB,TNF-α,IL-1β,and PAI-1 in the myocardial tissues of the ND group,ND+Pb group,and ND+Pb+P101 group.The results showed that,compared to the ND group,the expression of p-NF-κB,TNF-α,IL-1β,and PAI-1proteins in the ND+Pb group significantly increased(P<0.05);compared to the ND+Pb group,the expression of p-NF-κB,TNF-α,IL-1β,and PAI-1 proteins in the ND+Pb+P101 group significantly decreased(P<0.05).3.Compared with ND group,caspase-3 and p21 expression increased in myocardial tissue of ND+Pb group(P<0.001).However,compared with ND+Pb group,caspase-3 and p21 expression decreased in ND+Pb+P101 group(P<0.01).4.In the ND+Pb group,BNP expression in myocardial tissue was higher than in the ND group(P<0.001).In contrast,in the ND+Pb+P101 group,BNP expression was lower than in the ND+Pb group(P<0.01).4.Compared to the ND group,Western blot analysis of myocardial tissue in the ND+Pb group showed a significant increase in the expression of caspase-3,p21,and BNP(P<0.05).However,in the ND+Pb+P101 group,the expression of caspase-3,p21,and BNP significantly decreased compared to the ND+Pb group(P<0.05).5.Relative to the ND group,Western blot analysis of myocardial tissue in the ND+Pb group revealed a significant increase in GATA-3 expression(P<0.001)and a significant decrease in T-bet expression(P<0.01).Under the intervention of P101,GATA-3 expression significantly decreased compared to the ND+Pb group(P<0.001),while T-bet expression significantly increased(P<0.01).6.Upon analyzing IL-1βimmunohistochemically,it was observed that the ND+Pb group exhibited a substantial elevation in IL-1βexpression in comparison to the ND group.(P<0.05);however,in the ND+Pb+P101 group,the expression of IL-1βsignificantly decreased compared to the ND+Pb group(P<0.05).7.The immunoblotting results of Collagen1A1 protein showed that:compared with the ND group,the protein expression of Collagen1A1 increased in the ND+Pb group(P<0.01),while compared with the ND+Pb group,the protein expression of Collagen1A1 decreased in the ND+Pb+P101 group(P<0.05).8.Sirius Red staining analysis:In the ND+Pb group,the collagen fiber content was significantly higher than that in the ND group(P<0.01).However,in the ND+Pb+P101 group,the collagen fiber content significantly decreased compared to the ND+Pb group(P<0.05).At the same time,the immunohistochemical expression results of collagen 1A1 protein were consistent with the trends displayed by the Sirius Red staining analysis.Conclusion:1.Lead exposure can induce myocardial injury in mice2.Lactiplantibacillus plantarum P101 mitigates lead-induced myocardial injury and may be associated with TH1/TH2 balance. |
