Chronic obstructive pulmonary disease(COPD)is a disease characterized by narrowed airways and persistent airflow restriction.The disease is now the third leading cause of death in the world,causing increasing social burden and economic loss,and there is still a lack of efficient and obvious treatment methods.Physalis alkekengi L.,as a natural medicine with the same source of medicine and food,have been widely used in the treatment of lung diseases since ancient times.Scholars at home and abroad have found that Physalis alkekengi L.are rich in active compounds such as sterols and flavonoids,which have anti-inflammatory,blood-lipid-lowering,and anti-tumor effects.However,there are few studies on the treatment of COPD by Physalis alkekengi L.Therefore,this study aims to explore the effect of Physalis alkekengi L.alcohol extract on cigarette smoke through network pharmacology,metabolomics,and the construction of cell inflammation models and animal COPD models.Treatment effect and mechanism of action in chronic obstructive pulmonary disease.The specific research contents are as follows:1.In vitro experiment,the model of cell inflammation was established by smoke stimulation,and then the intervention therapy was given by Physalis alkekengi L.The results found that smoke stimulation affected the survival rate of A549 and enhanced the expression of the inflammatory factor TNF-α;40% concentration of Physalis alkekengi L.ethanol extract had no significant effect on cell survival rate,and could regulate the expression of intracellular TNF-α level.2.In vivo,an animal model of COPD was established in Balb/C mice smoked by cigarettes for 12 weeks.The results found that the alcohol extract of Physalis alkekengi L.improved the weight loss of mice caused by smoke,and the amount of food and defecation recovered after administration.And it can improve the physiological indicators of lung function such as Cdyn and RI in mice,repair the structure of bronchi and alveoli in lung tissue,and reduce the expression level of inflammatory factor TNF-α in lung tissue of mice.3.Network pharmacology analyse the main active constituents of Physalis alkekengi L.and its mechanism of action against COPD.It was found that the regulation of inflammatory response is one of the main mechanisms for Physalis alkekengi L.to exert its anti-COPD effect.The key target pathway maybe PI3K-Akt signaling pathway.4.Metabolomics analysis was performed using mouse serum.It found that linoleic acid metabolism,glycerophospholipid metabolism,sphingolipid metabolism,sulfur metabolism and tryptophan metabolism are related to the protective effect of the ethanol extract of Physalis alkekengi L.on COPD.In summary,Physalis alkekengi L.have certain anti-inflammatory effects,and may exert their protective effects on smoke-induced cell inflammation and COPD in mice by regulating the expression of PI3K-Akt and regulating metabolites in vivo.The above research results provide a theoretical basis for the development of new antiCOPD drugs.
