Objective : Based on endometriosis patients’ endometriosis(KPNA2 and HIF-1α)expressions,the relationship between them and clinicopathological parameters of endometriosis is analyzed,the correlation between the two is discussed,and the mechanism of action and clinical significance of them are preliminatively discussed.Methods : A total of 134 endometrial specimens were collected from the Department of Obstetrics and Gynecology of the First Affiliated Hospital of Nanchang University from April 2017 to October 2022 for concurrent surgical treatment,including 52 ectopic endometrium of ovarian endometriosis,31 present endometrium and 40 normal endometrium of non-endometriosis patients.All endometrial specimens were confirmed by pathology department of our hospital after surgery,and the clinical data were complete.According to different sources of specimens,they were divided into three groups: EMs ectopic endometrium group,EMs in situ endometrium group,and normal endometrium group.Immunohistochemistry was used to detect the expression of KPNA2 and HIF-1α in endometrial tissues of the three groups.Chi-square test was used to analyze the expression of KPNA2 and HIF-1α in endometrial tissues of the three groups and their correlation with clinicopathological parameters of patients with endometriosis.Spearman correlation analysis was used to detect the correlation between KPNA2 and HIF-1α expression in the ectopic endometrium and in the endometrium.Result:1.In patients with endometriosis,the expression levels of KPNA2 and HIF-1α proteins in ectopic endometrium were higher than those in endometrium and normal endometrium,and the difference was statistically significant(p < 0.05),the expressions of KPNA2 and HIF-1α in the endometrium were higher than those in the normal endometrium,the difference was statistically significant(p< 0.05);2.KPNA2 expression level was not correlated with the age of patients with endometriosis,endometrial cycle,ovarian endometriosis cyst(size and location),dysmenorrhea,recurrence,pregnancy number and abortion number,and the difference was not statistically significant(p > 0.05),was correlated with the clinical stage of ARSM,and the difference was statistically significant(p<0.05),HIF-1α was not associated with the above clinicopathologic parameters,and the difference was not statistically significant(p > 0.05);3.In the endometrial tissue,the expression level of KPNA2 protein in proliferating stage was higher than that in secreting stage,the difference was statistically significant(p < 0.05),but the expression level of HIF-1α protein was not correlated with the endometrial cycle;4.There is a significant positive correlation between the expression of KPNA2 and HIF-1α in ectopic and in-place endometrial tissues of endometriosis patients.Conclusion: KPNA2 and HIF-1a were highly expressed in both ectopic and in-place endometrium of patients with endometriosis,and showed a significant positive correlation,suggesting that KPNA2 and HIF-1α may be synergistic in the pathogenesis of endometriosis,but the specific mechanism of action needs further study.
