Background and purpose:IIFI30 is a reductase whose most recognized function is to enhance the process of MHCⅡ restricted antigens.Current studies have shown that IFI30 may be involved in the occurrence and progression of all kinds of malignant tumors including breast cancer and glioma.Nevertheless,its function in cervical cancer is remain unknown.Based on the expression of IFI30 in cervical cancer tissue samples,the study analyzed the connection between the expression level of IFI30 and the clinicopathologic features,survival data,tumor infiltrating cells,immune checkpoints and etc,in order to get new prognostic indicators and therapeutic targets for cervical cancer patients.Materials and methods:(1)we use some online databases(such as TIMER,GEPIA,HPA,UAICAN database)to comprehensively explore the expression level of IFI30 gene in cervical cancer tissues and its relationship with the prognosis of cervical cancer patients,immune cell infiltration,common immune checkpoint,etc;(2)Clinical information(such as clinicopathological features and follow-up data)of 78 patients with cervical squamous cell carcinoma was also analyzed retrospectivel,and Immunohistoche-mistry was used to detect IFI30 protein expression in tumor tissues of these cervical cancer patients and 20 normal cervical tissues.Then,we evaluated the correlation between IFI30 expression and clinicopathologic features and Overall survival of cervical squamous cell carcinoma patients.Results:The results of bioinformatics analysis showed that the expression level of IFI30 in cancer tissues was significantly higher than that in normal tissues,and the expression of IFI30 was different among patients with cervical cancer.When IFI30 was low expression,tumor infiltrating lymphocytopenia(including B cells,CD4+T cells,CD8+T cells,etc)and expression of common immune checkpoints(including PD-1,PDL1,CTLA-4,etc)decreased,and the patients with cervical cancer had shorter OS.Enrichment analysis suggested that the expression level of IFI30 was related to antigen presentation and treatment of NF-KB pathway.The IHC results showed that compared with normal cervical tissues,IFI30 protein expression levels in cervical squamous cell carcinoma tissues were significantly increased.K-M curve and multivariate COX proportional risk model analysis showed that IFI30 was an independent risk factor in patients with cervical cancer.Conclusions:1.Compared with normal cervical tissues,IFI30 is highly expressed in cervical cancer tissues,and there are differences among different cervical cancer patients:2.the expression of IFI30 is closely related to the immune microenvironment.3.Patients with low expression of IF130 have a worse prognosis,and low expression of IFI30 is an independent predictor of risk factors in patients with cervical cancer.
