| Periodontitis is an infectious disease caused by local factors and characterized by gingival inflammation and alveolar bone loss.The conventional treatment methods for periodontitis are plaque control,supragingival scaling,and subgingival scaling,along with adjuvant drug therapy.The most commonly used antibacterial drugs are ineffective in treatment and have toxic side effects,resulting in their inability to used for a long time.Therefore,searching for a new alternative therapy is the future research and development trend in the treatment of periodontitis.Numerous studies have shown that cryptochlorogenic acid has significant anti-inflammatory effects.However,the role and specific mechanism of cryptochlorogenic acid in periodontitis is still unclear.This study aims to explore the effect and mechanism of cryptochlorogenic acid on periodontitis,for clinical transformation and application.Objective: To explore the effect and mechanism of cryptochlorogenic acid in the treatment of periodontitis through the periodontitis model.Methods:1.CCK-8 detection of toxicity of different concentrations of cryptochlorogenic acid on RAW264.7 cells.2.Flow cytometry to detect the effect of cryptochlorogenic acid on the expression level of macrophages M1 polarization marker CD16/32.3.Transcriptome analysis of inflammatory gene expression changes after cryptochlorogenic acid intervention in the M1 polarization model of macrophages;q RT-PCR further validated the expression level of differential genes.4.Immunofluorescence staining and protein immunoblotting were used to detect the effect of cryptochlorogenic acid on the NF-κB/JMJD3 signal axis in the M1 polarization model of macrophages.Results:1.0 μM-20 μM cryptochlorogenic acid has little effect on RAW264.7 cell activity,and the cell survival rate is greater than 80%.2.Cryptochlorogenic acid can inhibit the expression of M1 polarization marker CD16/32.3.Transcriptomic analysis after cryptochlorogenic acid intervention in M1 polarization model,The results of KEGG enrichment analysis showed that the differential genes were enriched in synaptic vesicle cycle,aposi sarcoma-associated herpesvirus infection,long-term degradation,osteoclast differentiation,NF-κB signal path,and other signal paths.Sequencing results and q RT-PCR experimental results showed that cryptochlorogenic acid can inhibit the proinflammatory factor IL-1β And i NOS expression.4.Immunofluorescence staining and protein immunoblotting showed that cryptochlorogenic acid could inhibit NF-κB(p65)nuclear translocation and downregulation of JMJD3 expression.Conclusions: Cryptochlorogenic acid through NF-κB/JMJD3 signal axis regulation M1 polarization in the treatment of periodontitis. |
