| Objective Based on the role of specific inflammatory cell infiltration of lung tissue in pulmonary vascular reconstruction and the effect of blood viscosity on pulmonary artery pressure,the effect of Tetramethylpyrazine on pulmonary vascular peripheral inflammatory cell infiltration and the improvement of blood viscosity in hypoxic rats were studied,to explore the new mechanism of Tetramethylpyrazine in the prevention and treatment of hypoxic pulmonary hypertension.Methods 40 male SD rats were randomly divided into normal oxygen+normal saline group(CD),normal oxygen+ligustrazine group(CT),hypoxia+normal saline group(HD),hypoxia+ligustrazine group(HT).Rats in ligustrazine hypoxic exposure group were placed at a simulated altitude of 5000m in a hypoxic chamber for 28 consecutive days to establish a rat model of hypoxic pulmonary hypertension.Animals in the normal oxygen group were fed outside the chamber at the same time.The intragastric volume was 2mL,and the dosage of ligustrazine was 100mg/kg.The animal model was identified by monitoring the mean pulmonary artery pressure and right heart hypertrophy index,and the pulmonary vascular structure was observed by hematoxylin and eosin staining,and the influence of ligustrazine was analyzed by blood routine,blood biochemical and blood immune indexes.Arterial blood gas test oxygen saturation and oxygen partial pressure;The expression and distribution of CADM3 and CD11b+in lung tissues were detected by western blotting and immunohistochemistry.Results 1.After 28 days of exposure in low-pressure hypoxia chamber,right heart catheter method showed that compared with CD group,the mean pulmonary artery pressure of rats in HD group was significantly increased(P<0.05),which proved that the HPH model of rats was successfully replicated.2.After the intervention of ligustrazine,the mean pulmonary artery pressure and right heart hypertrophy index in HT group were significantly decreased compared with HD group(P<0.05).The results of hematoxylin and eosin staining of lung tissue showed that the thickening degree of pulmonary artery wall in the HT group was significantly reduced compared with that in the HD group(P<0.05),and the lumen diameter was increased.The results of arterial blood gas analysis showed that the arterial oxygen partial pressure(Pa O2)in HT group also increased compared with that in HD group.The results of blood routine and blood biochemistry showed that the mean red blood cell volume,distribution width of red blood cell volume and mean red blood cell volume of HT group were significantly decreased compared with HD group(P<0.05),and the mean hemoglobin concentration was significantly increased compared with model group(P<0.05).Conclusion Tetramethylpyrazine has a therapeutic effect on hypoxic pulmonary hypertension,and its mechanism may be related to reducing the infiltration of hypoxic pulmonary histitis cells and reducing blood viscosity,improving the oxygen-carrying capacity of red blood cells,and improving the hypoxia state of the body as a whole. |
