| Cornus officinalis Sieb.Et Zucc is the mature and dry pulp which was firstly recorded in Shennong’s herbal classic.The antioxidant,hypoglycemic,hypolipidemic,etc bioactivities of extracts were based on its abundant polysaccharides,iridoids,anthocyanins and triterpene.The safety and effectiveness of cornus had been affected less indicators,fuzzy origins and unclear changing composition.Hence,this study systematically evaluated the quality and processing mechanism of cornus.In addition,the basic properties and bioactive of cornus polysaccharides was also researched which promoted the effective utilization of cornus.In this paper,21 batches of cornus from five main production areas were studied by Chinese Pharmacopoeia.The results showed that the all samples were eligible which ensured real raw materials.Quality marker(Q-marker)is an important index for multi-component quantitative and fingerprints analyses which could be used to comprehensively evaluate the internal quality of traditional Chinese medicine.In this paper,5 chemical markers were screened by network pharmacology.Metabolomics revealed that18 differential components could be regarded as chemical markers.Identification of purified iridoid glycosides and polyphenols furtherly complemented the chemical markers.A stable HPLC method was developed to simultaneously quantity gallic acid,5-HMF,protocatechuic acid,morroniside,sweroside,loganin and cornuside I.The fingerprints and feedforward neural network model were establishd based on seven compounds to evaluate the quality and distinguish source of cornus.Bioactivity analysis furtherly pointed out that these seven components could be seen as Q-markers.Based on comparing chemical components between cornus and wine-prepared cornus,the transformation rule of 5-HMF,phenolic acid and iridoid glycosides at high-temperature were discussed.The results showed that the polysaccharide COP-2 was an acidic polysaccharide with a molecular weight of 16.13 k Da,which consisted of mannose,rhamnose,galacturonic acid,glucose,galactose and arabinose at 1.08:3.24:1.28:2.06:1:7.48 molar ratio.This results showd that COP-2 would be developed as hypoglycemic products.Above all,this paper evaluated the quality of 21 batches of cornus from five main production areas and established a comprehensive quality standard for cornus,.Meanwhile,the research of cornus polysaccharide could promote its development in food,medicine and other related fields. |
