Fomitopsis pinicola is a kind of edible and medicinal fungus with pharmacological activities such as anti-inflammatory,anti-tumor and antioxidant.At present,the extraction of F.pinicola only focuses on the effective extraction of its single functional component,while the extraction and utilization of other components can not be considered,resulting in the waste of resources and environmental pollution.Synchronous extraction design can improve the utilization of raw materials,obtain various types of bioactive components,reduce production costs and increase industrial economic added value.Therefore,this subject attempts to obtain two kinds of substances: Total Polyphenols(TP)and polysaccharides(PS)of F.pinicola by synchronous extraction technology at the same time.The activity of the extract is evaluated by in vitro and cell-level antioxidant experiments to verify the feasibility of the process,which has high theoretical significance and practical value for realizing the efficient utilization of a variety of active substances in edible and medicinal fungi.In this paper,the process of simultaneous extraction of TP and PS by hot water was discussed,and the physicochemical properties and antioxidant activities of these extracts were studied.Through the activated carbon decolorization experiment and alcohol precipitation treatment of the water extract,the optimum conditions for decolorization of polysaccharide extract of F.pinicola were obtained as follows: p H 3.5,temperature 40 ℃,amount of activated carbon 1.5%.Under these conditions,the comprehensive score of decolorization rate of polysaccharide solution was 81.26%.The evaluation results of antioxidant activity in vitro showed that when the concentration was 5 mg/m L,the polyphenols with alcohol extraction concentration of 40% had the highest scavenging rate of DPPH free radical,and the scavenging rate of DPPH free radical was 91.44%;The polyphenols with 70% alcohol concentration had the highest free radical scavenging rate of ABTS,and the free radical scavenging rate of ABTS was 98.96%;Polyphenols with 40%alcohol concentration had the highest hydroxyl radical scavenging rate,and the hydroxyl radical scavenging rate was 97.22%;The polyphenols with 60% alcohol concentration have the strongest reducing power,and the reducing power is 0.515.The results showed that the protective effect of TP on yeast cells damaged by UV was the best.When the concentration was 20 mg/m L,the survival rate of yeast cells was 48.64%;TP can improve the activities of SOD antioxidant enzymes and CAT antioxidant enzymes.At 20 mg/ml,the activity of SOD antioxidant enzymes was 450.43 μ/mg,CAT antioxidant enzyme activity was 348.43μ/g FW;PS had the best inhibitory effect on the content of malondialdehyde.At 20 mg/m L,the content of malondialdehyde was 0.038 μmol/L.The simultaneous extraction technology was used to separate the TP and PS active components from F.pinicola.The process was feasible,and the extract had strong antioxidant activity.These results provided a reference basis for the efficient utilization of F.pinicola.
