| Objective:To investigate the expression of SUMO1,SUMO2 and SUMO3 in triple-negative breast cancer tissues and its effect on the prognosis of patients.Methods:We downloaded a total of 603 cases of triple-negative breast cancer tissue and normal tissue information from the TCGA(The Cancer Genome Atlas)and GEO(Gene Expression Omnibus)database.R language software was then applied to perform quality analysis and differential genetic screening of the chip data from the GEO dataset and TCGA database,and collated the chip data for transformation and data processing.224 of patients diagnosed with primary triple negative breast cancer(TNBC)from Fujian Medical University Union Hospital in 2015-2017 were enrolled.Clinical data of patients were collected.The expression of SUMO1,SUMO2 and SUMO3 in triple-negative breast cancer tissue was detected by immunohistochemical staining.Correlations between SUMO1/2/3 expression and clinicopathological characteristics were identified by the chi-squared test.DFS and OS were calculated by the Kaplan-Meier method and differences between groups were examined with the log-rank test.Univariate and multivariate cox regression analysis was performed to evaluate SUMO1,SUMO2,SUMO3,age,tumor size,lymph node metastasis,histological grade and lymph vascular invasion on the prognosis of TNBC patients.Results:The data of 412 triple-negative breast cancer and 191 non-tumor breast tissues in GEO dataset and TCGA database were calculated by R language.The results showed that the m RNA expressions of SUMO1,SUMO2 and SUMO3 in triple-negative breast cancer tissues were significantly increased.Immunohistochemical staining method also indicated that the protein level of SUMO1/2/3 was significantly up-regulated in TNBC tumor tissues compared to the paired adjacent normal breast tissues.No associations between SUMO1/2/3 expression and clinicopathological features were observed.Univariate Cox regression analysis showed that tumor size,tumor histological grade,lymph node metastasis,lymph vascular invasion,high expression of SUMO1,SUMO2 and SUMO3 were correlated with disease-free survival(DFS)of triple negative breast cancer(P<0.05).Lymph node metastasis,high expression of SUMO1,SUMO2 and SUMO3 were correlated with overall survival time(OS)(P<0.05).Multivariate survival analysis showed that lymph node metastasis,tumor histological grade,high expression of SUMO1,and high expression of SUMO3 were independent influencing factors of disease-free survival(DFS)in triple-negative breast cancer patients(P<0.05).While the high expression of SUMO1 was identified as an independent factor for overall survival(OS)of patients with triple-negative breast cancer(P<0.05).Conclusion:The m RNA expression levels of SUMO1,SUMO2,SUMO3 in triple-negative breast cancer tissue were significantly increased in the TCGA database and GEO dataset compared to para-cancer normal breast tissues.The protein expression of SUMO1,SUMO2,SUMO3 was significantly higher in triple-negative breast cancer tissues than para-cancer normal breast tissues.In addition,high expression of SUMO1 and SUMO3 was a risk factor for TNBC tumor recurrence or metastasis.Elevated expression of SUMO1 was a risk factor for overall survival in patients with TNBC.SUMO1 and SUMO3 might be an important prognostic index for triple-negative breast cancer. |
