| Morinda officinalis How(Rubiaceae family),which is one of the “top four southern traditional Chinese medicines”,is mainly distributed in provinces of Guangdong,Guangxi,Fujian,Hainan,etc of China.The phytochemical investigations of M.officinalis led to the isolation and identification of various secondary metabolites including anthraquinones,naphthoate derivatives,iridoids and triterpenoids.Some of them displayed a series of biological activities,such as anti-depressant,anti-osteoporosis,anti-inflammation,anti-Alzheimer’s disease,anti-fatigue,anti-aging,and so on.In order to comprehensively elucidate the material basis of M.officinalis,we carried out a systematical phytochemical investigation on the dried roots of this plant.Thirty six compounds were isolated through TLC,silica gel,ODS,Sephadex LH-20 and HPLC chromatographies and their structures were elucidated on the basis of comprehensive spectroscopic.These compounds were identified and named as:twenty-two iridoid glycosides: morofficinaliside A(B-1),morindalin A(B-2),apodantheroside(B-3),monotropein(B-4),11-demethyl-apodanthoside(B-5),10-methoxy-11-demethyl-apodanthoside(B-6),morofficinaliside B(B-7),geniposidic acid(B-8),deacetylasperulosidic acid(B-9),scandoside(B-10),6-methoxygeniposidic acid(B-11),6β-ethoxygeniposidic acid(B-12),rhodolatouside B(B-13),6-O-acetylscandoside(B-14),deacetylasperuloside(B-15),asperulosidic acid(B-16),ixoside(B-17),forsythide(B-18),morofficinaloside(B-19),morofficinaliside C(B-20),secoxyloganin(B-21),morindalin C(B-22);nine anthraquinones: 2-hydroxy-1-methoxy-anthraquinone(B-23),1,3-dihydroxy-2-methyl-anthraquinone(B-24),3-hydroxy-1-methoxy-2-methyl-anthraquinone(B-25),1,2-dimethoxy-3-hydroxy-anthraquinone(B-26),1,3-dihydroxy-2-methoxymethyl-anthraquinone(B-27),1,3-dihydroxy-2-ethoxymethyl-anthraquinone(B-28),2,8-dihydroxy-1-methoxy-anthraquinone(B-29),2,5,8-trihydroxy-1-methoxyanthraquinone(B-30),1,3,8-trihydroxy-2-methoxy-anthraquinone(B-31);and five other constituents: vomifoliol 3’-O-β-D-apiofuranosyl-(1 → 6)-β-D-glucopyranoside(B-32),(1’R,3’R,5’R,8’S)-epi-dihydrophaseic acid β-D-glucoside(B-33),scopoletin(B-34),syringic acid(B-35),vanillic acid(B-36).Among of them,B-1,B-7,B-12 and B-20 are four new compounds.This study not only enriched the material basis of M.officinalis,but also provided a scientific basis for the further development and utilization of M.officinalis. |
