| Background:Liver cancer is the most common liver malignant tumor.Hepatocellular Carcinoma(HCC)accounts for about 90% of liver cancers.In my country,the 5-year survival rate is less than 15%,while in the United States it is only 18%.In recent years,the induction of iron death has become a promising alternative treatment for triggering the death of cancer cells,especially in those malignant tumors that are resistant to traditional treatments and have poor efficacy,Iron death plays a vital role in HCC.Therefore,in-depth exploration of the prognostic value of iron deathrelated genes for HCC has important clinical value to help clarify the early diagnosis and treatment of HCC,especially to improve the treatment strategy of HCC,and to improve precise individualized treatment and precise prognosis.Objective:(1)to screen out core genes related to iron death in HCC,Based on this,HCC prognostic model was established.(2)The study also analyzed the core genes and related enrichment pathways in detail,and explored the relationship between the iron death pathway and other potential pathways in HCC,to improve the early diagnosis and treatment of HCC.(3)To study the hub gene and the sensitivity analysis of tumor chemotherapy drugs,and guide the drug use.Methods:Download HCC transcriptome and clinical information from TCGA public database.Through differential gene selection combined with single factor analysis,core prognostic genes were screened out.Then through LASSO analysis,the final factors for constructing a prognostic model are obtained.Under the biological enrichment analysis of GO and KEGG,the potential mechanism in HCC is further explored.In this study,ss GSEA analysis was also carried out to seek the potential immune mechanism in HCC.Results:G6PD,HMOX1,MT1 G,and NQO1 were finally determined as the factors for constructing the model.The AUC curve of the prognostic model shows a sensitivity of 0.679 at 1 year,0.660 at 2 years,and 0.617 at 3 years.Univariate and multivariate Cox regression analysis showed that the risk score was an independent prognostic risk factor,and the survival analysis based on the risk score showed that high risk had poor survival time(P=0.0036).The enrichment analysis of GO and KEGG showed that not only iron death-related collaterals were enriched,but also immunerelated pathways were also enriched.ss GSEA analysis showed that some immune cell scores and immune pathway enrichment were different between high and low risk groups(adj.P<0.05).Conclusion:The core genes found in this study were G6 PD,HMOX1,MT1 G,and NQO1.The prognosis model based on iron death-related genes in this study has a certain predictive ability and can predict the prognosis of HCC.In HCC,the iron death mechanism is inherently related to the immune mechanism,which can have a certain prompting effect on the change of the treatment strategy of HCC. |
