| BACKGROUND Gastric cancer(GC)Gastric cancer is one of the cancers with the highest incidence in the world,and its morbidity and mortality are also very high in China.Invasion and migration of gastric cancer are important factors affecting the mortality of gastric cancer.HPSE is highly expressed in gastric cancer,which is closely related to the invasion and migration of gastric cancer.However,its mechanism has not been fully studied.Adhesion spot kinase(FAK)is involved in the progression of many cancers,and FAK has an effect on different tumor cells and different stages of cancer development.Based on i TRAQ technique and bioassay,it was found that the expression of FAK in human gastric cancer SGC7901 cells is highly correlated with HPSE,and may be involved in HPSE’s promotion of invasion and metastasis of gastric cancer.Adhesion function plays an important role in the process of invasion and migration.Objective:This study was to investigate the relationship between HPSE and FAK in human gastric cancer SGC7901 cells,and whether FAK participates in HPSE promoting the migration and adhesion of human gastric cancer SGC7901 cells.To provide basis for elucidating the mechanism of HPSE promoting invasion and migration of gastric cancer.Methods: 1.Lentiviral vector(LV-HPSE-RNAi)that interferes with heparinase gene(HPSE)and negative control virus were respectively transferred into SGC7901 cells by si RNA interference technique,and stable strains were screened by purinomycin.2.Western blot and q-PCR were used to verify the expression of HPSE and the expression of corresponding FAK.3.The scratch test and adhesion test were carried out to verify the change of cell migration and adhesion ability.4.si RNA interference technique was used to reduce the expression of FAK in heparinase silenced cells,Western blot and q-PCR were used to verify the expression of FAK,and scratches tests and adhesion tests were used to verify the effect of FAK on cell migration and adhesion.Results:1.Western blot and q-PCR demonstrated that si RNA successfully silenced HPSE expression in SGC7901 cells(P<0.05).2.Western blot and q-PCR results showed that FAK was up-regulated in HPSE-silenced SGC7901 cells,and the difference was statistically significant(P<0.05).3.Compared with SGC7901 cells and SGC7901 cells transfected with negative control virus,the migration and adhesion ability of SGC7901 cells HPSE silenced was decreased,and the difference was statistically significant(P<0.05).4.Western blot and q-PCR demonstrated that si RNA successfully reduced the expression of FAK in HPSE silenced SGC7901 cells,and the down-regulation of FAK could promote the migration and adhesion ability of cell.Conclusion:HPSE can promote the growth and migration of human gastric cancer SGC7901 cells and increase cell adhesion ability.HPSE can down-regulate the expression of FAK in human gastric cancer SGC7901 cells.HPSE may promote the migration and adhesion ability of human gastric cancer SGC7901 cells by down-regulating the expression of FAK. |
