| Objective About 15% to 25% of breast cancers are Triple Negative Breast Neoplasms(TNBC),accounting for 23.8% of the incidence of breast cancer in China.The main treatments for TNBC are surgery and chemotherapy.Due to the lack of indications of endocrine targeted therapy,there are still no valuable drugs in endocrine therapy and biotherapy now.By studying the specific expression of PD-1,PD-L1 and SOX2 in TNBC,this paper explored and analyzed the expression of these three genes in TNBC and the relationship between the corresponding clinicopathological features,so as to produce rationale for the diagnosis,treatment and prognosis of TNBC.Methods 1.Collect and screen case materials and specimens: The clinical case data of 49 patients with TNBC were collected,including age,tumor size,lymph node metastasis,histological grade,and TNM stage.2.Expression of PD-1,PD-L1 and SOX2 in TNBC: 49 paraffin embedded tissues of cancer and normal tissues around from the TNBC patients were sorted out and checked.After sectioning,the expression results of PD-1,PD-L1 and SOX2 genes in cancer tissues and normal tissues around cancer relied on the determination of immunohistochemical staining.3.Analyze the relationship between the expression of PD-1,PD-L1,SOX2 genes and clinicopathological features and the correlation between factors :Through SPSS22.0 statistical software,we analyzed the relationship between the specific expression of PD-1,PD-L1 and SOX2 and the patient’s age,tumor size,lymph node metastasis,histological grade and TNM stage,as well as the correlation between PD-L1 and Sox2 expression indexes.Results 1.Detected by immunohistochemistry,the positive expressions of PD-1,PD-L1 and SOX2 in TNBC were 36.73%、48.98%、69.38%,respectively,while the positive expressions of PD-1,PD-L1 and Sox2 in normal breast tissues around cancer were 12.24% 、 18.37% 、10.20%,respectively(P < 0.05).During the progression of normal breast tissue to TNBC,the expressions of PD-1,pd-11 and SOX2 increased.2.It was showed by the analysis of the relationship between the expression of PD-1,PD-L1 and SOX2 and the clinicopathological features of TNBC patients that lymph node metastasis and histological grade trended to increasing along with the expression of PD-1 in TNBC(P <0.05),but not age,tumor size and TNM stage(P > 0.05);lymph node metastasis,histological grade and TNM stage trended to increasing along with the expression of PD-L1 in TNBC(P <0.05),but not age and tumor size(P > 0.05);lymph node metastasis,histological grade and TNM stage trended to increasing along with the expression of SOX2 in TNBC(P < 0.05),but not age and tumor size(P > 0.05).3.Spearman correlation analysis was performed on the expression of PD-L1 and SOX2 in TNBC tissues(P < 0.01).It holds that there is a positive correlation between the expression of PD-L1 and SOX2.Conclusion 1.PD-1,PD-L1 and SOX2 are differentially expressed in TNBC tissues compared with normal tissues.And these can provide a new combined antibody basis for the diagnosis,treatment and prognosis of TNBC.2.After analysing the expression of PD-1,PD-L1,SOX2 and clinicopathological characteristics of TNBC patients,it shows that the expression of PD-1 is related to lymph node metastasis and histological grade of TNBC patients,and the expression of PD-L1 and SOX2 in TNBC is correlated with lymph node metastasis,histological grade and TNM stage.Which means the abnormal expressions of the three genes are related to the occurrence、development、invasion and metastasis of TNBC and suggests that their positive expressions in TNBC are related to poor prognosis.3.The study shows that the expression of PD-L1 and SOX2 is positively interrelated,which illustrates that they take synergistic effect on the occurrence and development of TNBC.Therefore,combined detection of the expression of PD-L1 and SOX2 can provide more comprehensive treatment guidance and provide a new combined therapeutic target for immunotherapy of patients with TNBC. |
