Objectives Bioinformatics and immunohistochemistry were using to detect CMTM4 gene in cancer and normal tissues of patients with rectal cancer.In this study researchers analyzed the expression of CMTM4 and the correlation with various clinicopathological features,explored the clinical significance of CMTM4 gene in rectal cancer,and evaluated its potential as an effective predictor of prognosis of patients with rectal cancer.Methods Information from different public databases(GEPIA2 database,Human Protein Atlas database,TIMER database,STRING database,Kaplan Meier Plotter database,TISIDB database and c Bio Portal database)was sorted and analyzed to put forward relevant assumptions.Immunohistochemical assays were used to analyze and verify the expression of CMTM4 in rectal cancer tissues and adjacent normal tissues by using gene microarray(75 cases of rectal cancer tissue microarray and 75 cases of adjacent tissue microarray),and the results were statistically analyzed.Clinicopathological factors were analyzed and studied concurrently.Results 1 The results of GEPIA2 database and Human Protein Atlas database showed that the expression level of CMTM4 in rectal cancer was higher than that in normal tissues.2 Although the survival analysis of Kaplan Meier Plotter database showed that the high expression and low expression of CMTM4 had no significant statistical significance in affecting the overall survival of patients with rectal cancer,it was verified on Human Protein Atlas database that patients with rectal cancer with high expression of CMTM4 had a better prognosis(P< 0.05).3 STRING database analysis showed that CMTM4 had obvious interaction with CMTM3,CKLF,CMTM2 and CMTM1,and the correlation scores were 0.938,0.909,0.909 and 0.909 respectively.4 c Bio Portal database analysis showed that the gene mutation rate of CMTM4 in various subtypes of colorectal cancer ranged from 0.3% to 3.57%,and the gene mutation frequency of CMTM4,CMTM3,CKLF,CMTM2 and CMTM1 in colorectal cancer ranged from 0.4% to 1.5%(CMTM4:0.6%;CMTM3: 1.5%;CMTM2: 0.6%;CKLF: 0.4%;CMTM1: 1.5%);CMTM4 is rarely mutated in patients with rectal cancer.5 TIMER database analysis showed that the expression of CMTM4 was positively correlated with B cells and CD8 + T cells,but not with tumor purity,CD4 + T cells,macrophages,neutrophils and dendritic cells.6 TISIDB database analysis shows that there is a certain potential relationship between CMTM4 and anti-tumor immune factors,and there are different close relationships between CMTM4 and various tumor infiltrating cells and immune regulatory factors.Immunohistochemical results showed that CMTM4 was positively expressed in cytoplasm,and the expression level of CMTM4 protein in rectal adenocarcinoma was significantly higher than that in adjacent tissues(P < 0.001).The expression of CMTM4 was significantly correlated with distant metastasis(P < 0.001).Conclusions 1 The expression of CMTM4 is higher in rectal cancer and lower in normal tissues.2 CMTM4 was significantly correlated with distant metastasis of rectal cancer.3CMTM4 can be used as a better diagnostic marker of rectal cancer,and CMTM4 is likely to play a role as a tumor suppressor gene in rectal cancer.4 CMTM4 has different close relationships with various tumor infiltrating cells and immunomodulatory factors.Figure16;Table2;Reference 80... |
