Effect Of Huayu Qushi Recipe On VEGFA And IL-6 Related To Iron Death Of KOA In The Early And Middle Stages

Objective: To investigate the mechanisation of Huayu Qushi Formula in treating iron death of KOA and its effect on related genes VEGFA and IL-6.Methods:(1)Use the String PPI website to build the PPI network diagram,and use five different algorithms,namely,Cytoscape plug-in proximity,Ec Centricity,EPC,MCC,and MNC,to further screen the three key genes.The expression of GSE1919 and GSE55235 in KOA was further verified by two data sets;(2)Collect prophase and intermediate KOA patients from the orthopedics department of our hospital from February 2022 to December 2022,according to the relevant inclusion criteria and exclusion criteria,and obtain the informed consent of the patients.The included samples were divided into the experimental group of 25 cases and the contrast group of 24 cases,and the relevant basic information of the patients was collected.The test group was treated with the prescription for removing blood stasis and removing dampness,and the control group was treated with iricoxib.The expression of VEGFA and IL-6 in the two groups was detected by ELSIA,and the VAS score,WOMAC score and clinical symptom score of TCM were obtained.Results: Bioinformatics: screening the prescription for removing blood stasis and dampness,iron death and KOA key genes(VEGFA,IL-6);(2)Clinical study: The VAS score,WOMAC score and TCM clinical symptom score of the test group and the control group were not significantly different before treatment and were comparable.After treatment,there was no statistical difference between the groups,but there was significant statistical difference between the groups before and after treatment.Both Huayu Qushi Decoction and Irecoxib could improve the VAS score,WOMAC score and clinical symptom score of KOA patients;There was no statistical difference in the related scores between the two groups after treatment;There was no statistical difference between the test group and the control group before treatment in terms of VEGFA and IL-6.Comparison between the groups after treatment:there are statistical differences,suggesting that both Huayu Qushi Decoction and Arecoxib can inhibit the expression of VEGFA and IL-6 in OA,while there are also significant statistical differences between the test group and the control group after treatment,indicating that Huayu Qushi Decoction has stronger effect on inhibiting VEGFA and IL-6 than Arecoxib.Conclusions: Huayu Qushi Formula can effectively treat KOA,and can affect the expression of iron death related factors VEGFA and IL-6,providing a new strategy for the treatment of KOA from the iron death pathway.
