Background and Objective: Renal cell carcinoma(RCC)is a relatively common tumor in the urinary system.Renal clear cell carcinoma(RCC)is the most common pathological subtype of RCC and one of the most common human malignant tumors.In recent years,the morbidity and mortality of renal clear cell carcinoma have been increasing year by year and showing a trend of younger age,which is a serious threat to human health.For patients with early renal cancer,surgery is generally feasible,but there is a certain recurrence rate.Although with the continuous development of medicine,more and more targeted drugs and immunotherapy have played an important role in the treatment of renal cancer,the prognosis of patients with middle and advanced renal cancer is still not ideal due to the unclear pathogenesis of renal cancer and the insensitivity of renal cancer to radiotherapy and chemotherapy.Therefore,we urgently need to find biomarkers that can guide the diagnosis,prognosis and treatment of kidney cancer.At present,there have been several studies reported that the expression of family 72 members A(FAM72A)with similar characteristics is significantly different in multiple cancer tissues and adjacent tissues,which may promote the progression of cancer.However,there have been few reports on the relationship between FAM72 A and renal clear cell carcinoma.In this study,bioinformatics methods were used to explore the expression of FAM72 A in renal clear cell carcinoma and its prognostic significance,as well as its related role with immune cells.Method:Transcripts of renal clear cell carcinoma and related clinical data were downloaded from The Cancer Genome Atlas database(TCGA,thecancer Genome Atlas),and the differential expression of FAM72 A in renal clear cell carcinoma and FAM72 A among different clinicopathologic feature groups were analyzed by R software.Using R software(KM,Kaplan-Meier Plotter)survival curve drawing,the receiver operating characteristics(ROC,receiveroperatingcharacteristiccurve)curve,by single factor,Multivariate COX regression analysis was performed to evaluate the independent prognostic factors of renal clear cell carcinoma,and a prognostic profile was constructed to evaluate the prognosis of renal clear cell carcinoma patients.gene aggregation enrichment analysis(GSEA)was used to explore the signaling pathways and potential molecular mechanisms related to FAM72 A.Through the TIMER(Tumor Immune Estimation Resource)database and R software analyzes FAM72 A relevance to immune cells infiltration and evaluated the related points of the tumor microenvironment,including matrix rating,immune scores and comprehensive scores of the two.Finally,R software was used to analyze the correlation between the expression level of FAM72 Am RNA and drug sensitivity.Results: The analysis showed that the expression of FAM72 A was up-regulated in renal clear cell carcinoma tissues(p < 0.05),and the higher the expression of FAM72 A,the lower the overall survival rate of renal clear cell carcinoma patients.Univariate and multivariate analysis found that age,Stage stage,Grade grade,and high FAM72 A expression were all factors affecting the prognosis of patients,and a Normogram was constructed to evaluate the prognosis.GSEA analysis showed that FAM72 A was closely related to homologous recombination signaling pathway,cell cycle signaling pathway and other signaling pathways.In the further immunoinfiltration analysis,it was found that the expression of FAM72 A in renal clear cell carcinoma was positively correlated with the infiltration of plasma cells,M1 macrophages,CD8+T cells and other immune cells(p <0.05),and negatively correlated with the infiltration of NK cells,dendritic cells,monocytes and other immune cells(p < 0.05).Conclusion: This study shows that FAM72 A is significantly highly expressed in renal clear cell carcinoma,and affects the prognosis of patients with renal clear cell carcinoma.Fam72 a can be used as an independent prognostic factor in patients with renal clear cell carcinoma,become a biomarker of renal clear cell carcinoma,and provide a potential target for immunotherapy of renal clear cell carcinoma.
