Objective: By treating human ovarian cancer SKOV3 cells with EGF-modified micellar formulations of paclitaxel and emoflavin,the absorption and uptake ability of SKOV3 cells to micellar formulations and its influence on the proliferation,invasion and adhesion ability of SKOV3 cells were observed,and the inhibitory effect of different micellar formulations on ovarian cancer was explored,in order to provide theoretical basis for future clinical application.Methods: In this study,ovarian cancer SKOV3 cells were cultured,taxol and emotin were coated in different micelles by thin film dispersion method,targeted molecules of Epidermal growth factor(EGF)were used to load on the surface of micelles,and compound formulas were prepared according to different ratios of EGF.FACScan flow cytometry was used to evaluate the uptake of different micellar formulations by SKOV3 cells using coumarin as a fluorescent probe.Paclitaxel and rhein were prepared into a variety of micellar formulations at different doses,and SKOV3 cells were cultured with a variety of micellar formulations.Cell survival rate was calculated by SRB staining,cell migration ability was detected by Transwell assay,and cell survival rate and adhesion rate were calculated.The inhibitory effects of different micelles on the proliferation,invasion and adhesion of SKOV3 cells were studied,thus confirming the effective targeting ability of drug micelles by EGF modification for ovarian cancer therapy.The data were presented as mean ± standard deviation(SD).Analysis of variance was used to calculate the significance between groups.All data were analyzed using SPSS20.0 software(social science statistical package),and P<0.05 meant the difference was statistically significant.Results:1.EGF-modified micelles can significantly improve the drug absorption capacity of SKOV3 cells.2.With the increasing proportion of EGF in the formula,the uptake capacity of SKOV3 cells to micellar preparation also increased.3.Paclitaxel significantly inhibited the proliferation of SKOV3 cells,and the inhibitory effect on SKOV3 cells was directly proportional to the dose.4.The sequence of inhibitory effects of various free drug formulations on SKOV3 cell proliferation was as follows: free taxel + emodin(1:10)> free taxel + emodin(1:5)> free taxel + emodin(1: 1)> free paclitaxel > free emodin.It can be seen that although emodin has no lethal effect on SKOV3 cells,the toxicity of paclitaxel on SKOV3 cells increases with the addition of emodin concentration ratio,indicating that emodin increases the inhibitory effect of paclitaxel on SKOV3 cell proliferation.5.The inhibitory effects of various micellar formulations on SKOV3 cell proliferation were as follows: EGF-modified taxol micelle + EGF-modified emodin micelle > taxol micelle + emodin micelle > taxol micelle > emodin micelle > blank control.These results indicated that the combination formula of EGF-modified paclitaxel micelles and EGF-modified emodin micelles enhanced the inhibitory effect on SKOV3 cell proliferation.6.The results of Transwell experiment showed the inhibitory effects of various micellar formulations on SKOV3 cell invasion: EGF-modified paclitaxel micelle + EGF-modified emodin micelle > paclitaxel micelle + emodin micelle > paclitaxel micelle > emodin micelle > no control,indicating that EGF-modified paclitaxel micelle plus EGF-modified emodin micelle enhanced the blocking of SKOV3 invasion in vitro.7.The inhibitory effects of various micellar formulations on SKOV3 cell adhesion were as follows: EGF-modified paclitaxel micelle + EGF-modified emodin micelle > paclitaxel micelle + emodin micelle > emodin micelle > paclitaxel micelle > EGF-modified emodin micelle > no control,indicating that EGF-modified paclitaxel micelle + EGF-modified emodin micelle enhanced the blocking of SKOV3 adhesion in vitro.Conclusions:1.EGF-modified micelles can significantly improve the drug absorption capacity of SKOV3 cells.2.The inhibitory effect of paclitaxel on SKOV3 cell proliferation was directly proportional to the dose of paclitaxel.3.Emodin enhanced the inhibitory effect of paclitaxel on SKOV3 cell proliferation.4.EGF-modified paclitaxel micelles plus EGF-modified emodin micelles enhanced the inhibitory effect on SKOV3 cell proliferation,invasion and adhesion.
