Objective:1.To confirm that Rhizoma Drynariae can regulate the Pyroptosis with the pathway of NLRP3/GSDMD/Caspase-1 and improve postmenopausal osteoporosis.In the co-culture of the osteogenesis/adipogenic transwell system,increased lipotoxicity was accompanied by the involvement of the Pyroptosis.METHODS:The osteoporosis rat model(OVX group)was constructed and grouped into sham-operated group(SHAM)and Rhizoma Drynariae group(OVXDF).After drug administration,serum ELISA,micro-CT of femur,histopathology of femur and qRT-PCR were performed to detect the levels of cell scorching-related and osteogenic genes.The expression of NLRP3,GSDMD,Caspase-1,PPAR-y and RUNX2 proteins were detected by Western Blot during the co-culture period.RESULTS:Bone density decreased and expression of cell scorch death standards increased in the tissues of rats in the three OVX groups,while expression of cell scorch death related indexes decreased and osteoblasts expression increased in the OVXDF group;PPAR-y increased,RUNX2 expression decreased and expression of cell scorch death standards increased significantly with the prolongation of co-culture time in the Transwell co-culture system.Conclusion:Rhizoma Drynariae can reduce the expression of Pyroptosis related indexes and play an anti-osteoporosis effect.Increased lipotoxicity during osteogenesis/lipogenic co-culture with activation of Pyroptosis.
