ObjectivesTo study the spatiotemporal expression and significance of mesenchymal stem cell markers CD24,CD29,CD90 and CD133 in the kidney development of normal Kunming mice,It provides theoretical basis for kidney injury and repair at the level of kidney development.MethodsSelect healthy Kunming mice,2 months old,weighing 24-28 g,and feed them without padding according to the ratio of male and female 1:1.It is observed that the fall off of the vaginal plug is recorded as 0.5 days of embryonic age(E0.5 days),and the first day after birth is recorded as P1 days.According to the development period,the kidneys of E14.5d,E16.5d,P1 d,P3d,P5 d,P7d,P14 d and P28 d mice were taken respectively.The left kidney was made into paraffin sections,and the right kidney was made into frozen tissue samples for immunoblotting.Immunohistochemical staining was used to detect the expression of CD24,CD29,CD90 and CD133 in mouse kidney development.BCA was quantified in the right kidney tissue,and the above markers were detected by immunoblotting.In addition,the kidney tissue of Kunming mice from 1-5 days after birth was taken,and the cells were isolated by enzyme digestion for cell culture in vitro.The expression of the above markers in the adherent cells was determined by immunofluorescence detection.ResultsImmunohistochemical staining of the kidney of Kunming mice at different developmental stages showed that CD24 was expressed in ureteral buds,S bodies,comma bodies,glomerular cells and renal tubular epithelial cells at E14.5-P1 d,and weakened at the glomerulus at P3d-28 d,mainly in the proximal and distal tubular epithelial cells in the renal cortex,and also a small amount in the renal medulla;At E14.5-P3 d,CD29 was expressed in ureteral bud,S body,comma body,renal tubular endothelial cells,and at P5d-28 d,in proximal and distal tubular epithelial cells;CD90 was expressed in the posterior renal mesenchyme at E14.5d-E16.5d,and P1d-28 d in the proximal and distal tubular epithelial cells,but not in the glomerulus;CD133 was expressed in comma body,S body,posterior renal mesenchyme and ureteral bud at E14.5d-E16.5d,P1d-28 d in proximal tubule and distal tubule epithelial cells,and in collecting duct at renal papilla area.The results of immunoblotting showed that the expression of CD24 began to increase at E14.5d,reached the peak at P1 d,and then began to decline;The expression content of CD29 reached its peak at P7 d,and then decreased gradually;The expression content of CD90 reached its peak at P14 th day,and then decreased;The expression of CD133 reached the highest level at P3 d after birth,and then began to decline.On the 14 th day of primary cell culture,long fusiform cells appeared and gradually formed clonal growth.The above markers showed positive expression by immunocytochemical fluorescence detection.ConclusionsMesenchymal stem cell marker CD24,CD29,CD90 and CD133 have a certain time and space character in the process of kidney development.They affect the kidney development of Kunming mice,especially the differentiation and development of early renal corpuscles,renal tubules and collecting ducts.
