ObjectiveExplore the effects of ketamine on liver cell activity and NO,cGMP levels in rats with liver injury.MethodsForty normal rats were selected and evenly divided into a normal group,a model group,a ketamine group,and a propofol group,with 10 rats in each group.Except for the normal group,all other groups established liver injury rat models.After successful modeling,the propofol group was injected with 1.0ml/1dose/day of propofol,the ketamine group was injected with 0.5mg/1 dose/day of ketamine,and the normal and model groups were treated with equal doses of physiological saline,The morphology of hepatocytes of experimental rats in each group was observed by HE staining,the apoptosis of rats in each group was detected by TUNEL,and the expression of smooth muscle actin(a-SMA),cytokeratin(CK)protein was detected by Western blot.ResultsCompared with the normal group,the model group had higher levels of liver tissue plasma CIV,serum NO,and cGMP(P<0.05).Compared with the model group,the ketamine group had lower levels of these indicators(P<0.05),while the propofol group had higher levels of these indicators(P<0.05).The liver plate texture of the normal group experimental rats is clearly visible,and the liver cells are arranged neatly and orderly,with a clear cord-shaped structure;The model group of rats showed significant infiltration of inflammatory cells in the liver,with severe nuclear pyknosis visible within the liver cells,abundant fat droplets,unclear cord like structures,and disorderly arrangement of liver cells with narrow hepatic sinusoids;The liver plate texture of ketamine group experimental rats is clearly visible,inflammatory cells are rare,and the arrangement of various liver cells is relatively neat,and the cord like structure tends to be clear;The texture of the liver plate in the propofol group is slightly clear,and there is still swelling in liver cells,inflammatory cells,and the central vein is still dilated with fat droplets.Compared with the normal group,the apoptosis rate of liver cells in the model group rats increased(P<0.05).Compared with the model group,the apoptosis rate of liver cells in the ketamine group decreased(P<0.05).Compared with the ketamine group,the above indicators in the propofol group increased(P<0.05).Compared with the normal group,the model group rats had higher levels of a-SMA and CK protein(P<0.05).Compared with the model group,the ketamine group had lower levels of these indicators(P<0.05),while the propofol group had higher levels of these indicators(P<0.05).ConclusionsKetamine has a certain effect on reducing liver tissue homogenous CIV,serum NO,cGMP levels,and inhibiting liver cell apoptosis in liver injury rats.
