Aim:TRPV4 receptor and HIPPO pathway play an important role in the repair of peripheral nerve myelin sheath after injury.By studying the differences between the sciatic nerve myelin sheath of wild type(TrpV4+/+)diabetic mice and TRPV4 knockout(TrpV4-/-)diabetic mice confirm that TRPV4 knockout upregulates the expression of HIPPO pathway,which is beneficial to the repair of peripheral nerve myelin sheath in diabetic mice.Methods:The mice models of type I and type II diabetes mellitus were established by intraperitoneal injection of streptozotocin(STZ).The differences of behavior and myelin protein expression in sciatic nerve between TRPV4 knockout mice and normal mice in different types of diabetes were compared,and then researched the relationship between TRPV4 receptor and the expression level of YAP/TAZ,a co transcription factor of HIPPO pathway.Results:1.The sciatic nerve conduction velocity of TrpV4-/-diabetes mice was faster than that of TrpV4+/+diabetes mice;2.The pain threshold of TrpV4-/-diabetic mice in behavioral experiment was higher,indicating that the pain sensitivity caused by diabetes was reduced;3.TRPV4 receptor knockout increased the expression of myelin protein in diabetic mice;4.TRPV4 receptor knockout,the expression of YAP/TAZ co transcription factor was upregulated,which promoted the repair of peripheral nerve myelin sheath.Conclusion:TrpV4-/-can repair peripheral nerve demyelinating lesions in diabetic mice by upregulating the expression of HIPPO pathway.
