Objective: This test is to confirm whether the activation of the NLRP3 inflammasome by the endogenous neurotoxin SAL-induced pyroptosis is involved in the pathogenesis of Parkinson’s disease.Methods: Human neuroblastoma SH-SY5 Y cells were selected.This experiment first determined the neurotoxic effect of SAL on SH-SY5 Y cells.After treatment,cell viability was measured by CCK-8.RNA sequencing(RNAseq)and Kyoto Encyclopedia of Genes and Genomes(KEGG)analysis were used to detect differentially described genes in SAL-treated SH-SY5 Y cells.On the other hand,the expression of NLRP3-cell pyroptosis-related proteins was detected by the following methods: 1.SH-SY5 Y cells were simulated for 24 h with or without the addition of SAL(300 μM).The levels of the protein m RNA and protein in each group were detected by qRT-PCR and immunoblotting,respectively,and β-actin was used as a loading control.2.SH-SY5 Y cells were disposed with various consistence of SAL for 24 h,and the levels of pyroptosis-related proteins were determined by western blot.3.SH-SY5 Y cells were handled with various consistence of SAL for 24 h,and the levels of pyroptosis-related proteins were determined by immunofluorescence.The test results showed that: we first determined the neurotoxic effect of SAL on SH-SY5 Y cells.RNA sequencing and KEGG analysis was performed to detect the differentially described genes in SAL-treated SH-SY5 Y cells.We found that NLR family-containing pyrin domain 3(NLRP3)-dependent pyroptosis was enriched by SAL;SAL was found to significantly increase the expression of NLRP 3-dependent pyroptosis genes at the m RNA and protein levels in SH-SY5 Y cells.It is indicating that NLRP3-dependent pyroptosis functions in SAL-induced PD pathogenesis act on.Proptosis-dependent neurotoxicity is further highlighted by SAL as a potential therapeutic target for PD.
