stagnation and blood stasis with Xuefu Zhuyu Decoction,studies the mechanism of the effect of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation from three dimensions of its overall metabolism,lipid metabolism,and endogenous metabolism changes of platelets,and further carries out the non-target metabonomics research of the disassembled prescription of Xuefu Zhuyu Decoction,and excavates the characteristics of the metabonomics of the effect of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation.ObjectiveUsing three methods of plasma non-target metabonomics,lipomics and platelet metabonomics,the mechanism of the effect of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation was explained from three dimensions of overall metabolism,lipid metabolism and platelet endogenous metabolism.On this basis,the metabonomics study of Xuefu Zhuyu Decoction for regulating qi and activating blood circulation was carried out,and the characteristics of the efficacy grouping and compatibility of Xuefu Zhuyu Decoction were discussed,To provide scientific basis for the clinical efficacy of Xuefu Zhuyu Decoction and the principle of its formulation.MethodThis study is divided into two parts,which are based on non-target metabonomics of plasma,lipomics,and platelet metabonomics technology,the study of the mechanism of the effect of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation,and the study of the compatibility between the effects of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation based on non-target metabonomics of Xuefu Zhuyu Decoction(Taohong Siwu Decoction,Sini Powder).Part Ⅰ:Based on the study of the effect of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation in non-target metabolomics of plasma,the rats were divided into normal group,model group,and Xuefu Zhuyu Decoction group.The rats in the Xuefu Zhuyu Decoction group were given preventive medication one week in advance,and then the rats in the Xuefu Zhuyu Decoction group were given epinephrine for seven days,with a dose of 0.6mg/kg.The Xuefu Zhuyu Decoction group was given traditional Chinese medicine at the same time,and the materials were anesthetized one hour after the last injection of epinephrine,According to the general state,blood routine,blood biochemistry,blood coagulation and other indicators of rats,the therapeutic efficacy of Xuefu Zhuyu Decoction was confirmed.The non-target metabonomics technology,lipomics technology,and platelet metabonomics technology were used to conduct in-depth research on the efficacy of Xuefu Zhuyu Decoction in regulating qi and activating blood circulation,and to explore the key metabolic pathways and biomarkers for regulating the efficacy of Xuefu Zhuyu Decoction in regulating qi and activating blood circulation,The mechanism of the effect of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation was discussed comprehensively from the perspective of metabolic changes.Part Ⅱ:The plasma non-target metabonomics technology was used to carry out metabonomics research on Xuefu Zhuyu Decoction,Huoxuezi Decoction,Taohong Siwu Decoction and Liqizi Decoction.Sini Powder.From the pharmacological indicators and plasma non-target metabonomics results,the differences and similarities between the effects of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation and the compatibility characteristics were discussed.ResultsPart Ⅰ:Based on non-target metabonomics technology,lipomics technology and platelet metabonomics technology,the mechanism of the effect of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation was studied.The results showed that the key metabolites regulated by Xuefu Zhuyu Decoction mainly included nonferrous acid,taurine,glutamic acid,guanine,oleic acid,3b,12a-dihydroxy-5a-cholic acid,phosphatidylcholine,phosphatidylserine,ceramide and other biomarkers,The main metabolic pathways involved are the metabolism of tryptophan,glutamic acid,taurine,purine,bile acid,carnitine,fatty acid,glycerol and phospholipid,and sphingolipids.Classified by the main functions of metabolic pathways,these metabolic pathways mainly focus on amino acid metabolism,purine metabolism,lipid metabolism,energy metabolism,and involve important physiological activities such as platelet function regulation,neurotransmitter transmission,energy metabolism homeostasis,etc.Part Ⅱ:Through the study of plasma non-target metabonomics of Xuefu Zhuyu Decoction for regulating qi and activating blood circulation,the characteristics of grouping and compatibility of Xuefu Zhuyu Decoction for regulating qi and activating blood circulation were discussed.The results of plasma non-target metabonomics showed that the characteristics of Xuefu Zhuyu Decoction for regulating qi were reflected in the regulation of purine metabolism closely related to the emotional state of the body,and the characteristics of activating blood circulation were the metabolism of fatty acids directly affecting the function of blood platelets in the body The regulation of taurine metabolic pathway and the intervention of glycerol phospholipid metabolism are the key ways of regulating qi and activating blood circulation.After the compatibility of the effect of regulating qi and activating blood circulation,the regulation effect of Xuefu Zhuyu Decoction on the metabolism of tryptophan and glutamate in the body is significantly enhanced,while the regulation effect on the metabolism of bile acid and carnitine is weak,indicating that the efficacy compatibility may strengthen the effect of Xuefu Zhuyu Decoction on the intervention of platelet aggregation and neurotransmitter transmission,And weaken the regulation of energy metabolism in the body.ConclusionThe mechanism of the effect of Xuefu Zhuyu Decoction on regulating qi and activating blood circulation is mainly reflected in regulating the key metabolites such as tryptophan,taurine,glutamic acid,guanine,oleic acid,3b,12a-dihydroxy-5a-cholic acid,phosphatidylcholine,phosphatidylserine,ceramide,etc.in the body,regulating the important metabolic pathways such as amino acid metabolism,purine metabolism,lipid metabolism,energy metabolism,and interfering with the activation and aggregation of platelets in the body,energy metabolism homeostasis Neurotransmitter conduction and other important physiological processes.The results of plasma non-target metabolism showed that its effect of regulating the metabolism of purine and activating blood circulation was characterized by regulating the metabolism of fatty acid and taurine.The
